hobbit, have you tried doing a transfer to water agar? iโ€™ve had trouble with plates splitting like that despite a bunch of transfers. put them to water agar & have just recently taken a transfer back to a nutritious plate.  itโ€™s a bit too early for me to make definitive conclusions, but so far the growth on those transfers looks better. could be worth trying if youโ€™re just fucking around with the plates

edit: just noticed luminous mentioned water agar too

You know honestly itโ€™s not getting out to grain anytime soon regardless. Iโ€™m germing, cleaning, cloning, and storing my library of 40+ varieties. The line of already germed plates waiting to grow is pretty long. Iโ€™ll go back to spore.

What's the rush? Just let it go and show you where the knots will show up. I've seen cultures like that that are just fine.

More often than not those scalloped cultures are tricky as hell and often are very susceptible to mold infections later on although some of them kick ass. Too early to know.

Blue tinge on surface of otherwise healthy looking mycelium growth? [Re: Nichrome]
    #28051653 -

First time with this PE strain, from syringe I purchased.  This plate looks very much the same as three others inoculated.  Was completely white until today, when I noticed a lightly blue tinge on the top of the growth.

What do you all think? Is this within normal color variation or do I have a problem?

That a germ plate? Just take transfers. The blue might only be bruising.

I don't see any problems other than the disorganized pattern of a culture that hasn't decided who it is yet.

First successful clone.

Should I take a transfer now or let it grow out some? New to cloning on agar

I think it's been a week, maybe six days. Close enough. Pretty sure I've got some clean myc and as a result, I've been wondering -- when does one start to think about making transfers?

That brings me back to y'all for guidance! I've still not finished the light box so the photos are still meh, and for that I apologize. The condensation on the lids makes things look suspicious but what I can see in person, none of what's going on up on the lids is happening on the surface of the agar, for what that's worth. That condensation is a tricky bastard to contend with... Anyways.

First up is APE. 12 oclock looks interesting, but suspect given its position. Could've been from where the spore solution wandered while wrapping the plate in parafilm. Otherwise, looks like there are some good candidates to go onto T1 plates.

Next is psilocybe cyanescens. This one was trickier to get the admittedly crappy pics of due to the condensation.

My untrained eye doesn't see anything super concerning (aside from the hideous condensation on the lids), but that's very likely due to lack of experience. Any input on where/if either of these plates are viable and or ready for transfers would be awesome!

Decent germ plates. Make transfers pretty soon. You can get nice cultures from both of those.

Just found this at the bottom of a stack of old plates.

Iโ€™m doing grain prep today. I think Iโ€™ll toss this into a jar and see what happens.

If you're going to toss it in a jar, you should cut a piece off and toss it on a plate so you'll already have it on agar if the test jar does well
