certainly looks like it has potential. Let it grow out a little further and you'll have a better idea in a few days.

Best to learn patience now. Everything in this hobby is a few days away, a couple weeks until 'whatever'...unless you get busy and put off harvesting for another day then your entire crop is black with spores

haha yeah, been there. admittedly it's been over a decade since I've had a monotub blow its load due to college era shenanigans.

after waiting two weeks for the first round of plates to do nothing with one being confirmed contaminated, and now this batch taking five days to mostly do nothing, i'm kinda lacking in that dept now haha. BUT with confirmed "good" growth, i won't open the incubator for a few days and let it do it's thing uninterrupted.

even if this round ends in 100% contamination, success has been achieved! germination on agar is possible for a rusty old man like me. now the real work can begin :happiness:

it's just my poor photography skills that got ya!

GERMINATION Y'ALL!!!

Sorry for the excitement, my kids don't give a shit about microbiology yet, and I don't share this hobby with anyone else in my life, so you all have the privilege of dealing with my excitement here.

A couple lessons learned, I think.

First, I'm pretty sure my agar is too hard, that's why the first five plates I tried anything with did nothing. Did 10g agar, 10mg LME, 500ml and pre-boiled before PCing (learned on this thread that last step was a poor idea).

Second, pre-stretching the parafilm seems to be SOP. The parafilm has split on a handful of plates, so will be trying the pre-stretch next time.

Will also be trying 7g agar instead of 10g to 500ml water, see if softer agar works out better for germination.

Otherwise, here is a pic of an APE multispore plate I knocked up this past weekend. Of the three plates I hit with these spores, this is the only one that is showing signs of life. Also did three with psilocybe cyanescens, and three with pan cambo, and none of those have shown any signs of doing anything.

Just wanted to celebrate here a little and say thank you to all you fine people that have posted so much information, lessons learned, etc for decades. It's much appreciated! I'm working on a setup for better plate pics as well, so promise I'll quit dropping these bad ones here soon

Hey m8!

Don't worry, I share your excitement for germination ;-) Nice work!

Regarding Agar, I had the same feeling with those measurements, I work now with 8gr Agar, 8gr LME, (opt. 0.5g nut yeast) for 500ml and that seems for me to be just right ;-)

In my time in a Lab, I learned that Agar wont get boiled. Wen it gets boiled in the lab, they toss it. It's  enough when heating it on the stove until alot of steam rises but not yet boiling. You notice that at some point, the liquid starts to foam, that's when all the nutes and agar got dissolved. Then I fill it in a bottle to PC for 20min.

Regarding Parafilm, don't pre-strech it. If you do, it won't stick so nicely to the dishes which gives opportunity's for small gaps. Wen I wrap my dishes, I hold one end with my finger, strech about 1cm, hold again where I stopped with stretching, and continue.

Sometimes germination can take a couple of day's until you notice growth. Your agar recipe works, just give it some time until they germinate. My mexicana germinated after 6 days.

Looking forward to your updates!

Cheers & mush love <3

I’m like pretty sure that’s all bacteria or bacteria mostly barely mixed with mycelium. Kinda hard to tell and I may be wrong.

I did see the "foaming", though I'm not sure that's what I would call it. It was more like giant bubbles of gelatin, and I was sure I was going to have to be googling "how to get boiled over agar off every surface in a kitchen." In other words, I will definitely NOT being doing that again.

I was thinking it over last night, and I'll probably make up a few batches next round. Getting the recipe dialed in for one's self seems to be half the battle.

In the interim, I've dug out my old nikon DSLR and will be setting up a light box as to address the photography situation. That agar photography thread has some quality nuggets in it so it's something to do while impatiently waiting for progress!

How hard did that agar turn out? And isn't that pretty rich on the nutes? And if you're only PCing agar, 20 minutes at 15 psi should be enough, right everyone?

I honestly questioned that too, so I figured that was the most polite way to correct the record. I can see some newb looking for the latest and greatest agar recipe, finds and reads that, and immediately starts cooking. I just want to save someone the trouble and time.
