Re: AGAR ENVY! (Anything and All things agar!) [Re: Guerrilla]

Stack poured with the addition of nutritional yeast :bow2: Let's try and get some decent looking growth.

And my 2nd contaminated plate straight out of the stack in a year of pouring near enough 500 plates...

I only noticed it after I put the tissue sample on it. These were the half dose LME stack that needed using. I may have knocked that stack and caught it before it fell on the floor though. (Don't tell anyone!)

Grains from an empty, dried out 8 month old spawn jar I found and put to agar a week ago. Saw some myc jumping off one of the grains today. I'm SO freakin excited I didn't lose this culture for good!!!

Yea, and they are both people making their own prints and syringes and knowing what they’re doing. Not vendor syringes.

Yes it CAN work if you are running enough jars but majority of the time it won’t, so I personally don’t agree with setting a beginner up for failure. Not trying to be a dick they just might get disheartened with their almost guaranteed failure.

I know you suggested PF tek. I was just confirming that this was the more suitable option.

You're suggesting PF tek, but suggesting NOT injecting spore syringes into grain. PF tek IS injecting spore syringes into grain. (EDIT: Technically. Yeah, I know.)

I'm confused with your suggestions. And I bet the new guy is too by now.

FYI, I started by injecting vendor syringes into grain. (Not PF Tek). That gave allowed me to make my own spore prints and have more to work with.

I've since got better spores (prints and swabs) and gotten good at agar. But if I had to go from syringes to agar off the bat, I probably would have bailed. I was horrible at agar off the bat, even after growing some shrooms.

There's a lot to be said for someone new to this hobby to just be able to grow something...gives them hope to keep going ...to work on getting better, with more complex teks. Also gives them more genetic material than a spore syringe.

If you have better suggestions, maybe DM the guy and give him some step by step advice. Watching people bicker on here isn't helping him.

It’s brown rice flour and vermiculite...  much more resistant to bacterial contamination. It is not the same thing as inoculating standard grains. And if you think that then I’m honestly quite shocked.

Also, I am not bickering lol? Was simply stating facts, sorry to hear that’s what you’re doing.. bickering.. but I’m not on the shroomery to be arguing.. This isn’t the first time I’ve seen you trying to argue on the forum, seen it quite a few times now. The fact that in your short time here you are ignored by 3 members and I will now be your 4th says it all. This is a waste of energy on both parts and quite frankly it’s embarrassing. Nobody wants to see this shit, so I won’t be replying if you really feel it necessary to. Peace, love, you go on your merry way now.

I transferred some TED spores to agar and not sure if I should be concerned about the yellow?

Thanks!

So, can you tell us a little about your technique? Are you using a SAB (Still air box)? (If you can drop 500 to 800 bucks, a flow hood is a game changer, but all this can be done with a simple SAB. And most people here use an SAB. You can make one for 5 or 10 dollars).

Are you using ISO alcohol to spray your work area, inside your SAB, and your gloves? You should. Don't bother buying lysol, just get 70% (not 90%) Isopropyl alcohol and put it in those little spray bottles people use for laundry. Mark it clerarly, so you don't mix it up with your water spray bottle, which you'll want when you grow shrooms to mist dry substrate.

DO NOT use flame inside your SAB. Will start a fire.

Are you using flame to sterilize your scalpel? What is your flame source? If it's an alcohol lamp, I'd ditch it and get a little butane torch (like people use for brazing food when cooking, they're often sold as that on amazon and elsewhere.) Pic of mine in this post:
https://www.shroomery.org/forums/showflat.php/Number/27554328#27554328

Are you wearing disposable gloves (you should). Are you wearing a face mask? I recommend it. A few don't, but if you don't you're breathing on the agar. Doesn't have to be fancy, just a clean cloth mask works.

Are you following a particular tek? Which one?

Are you pouring your own plates? How are you sterlizing your agar mix, and what's in your agar mix?

Do you have a pressure cooker? What make /model, and how are you using it?

These are the things I can think of off my head that are vectors for contam, that before I changed them, I had plates like yours. Never yellow, but that contammed.

Let me know and I'll see if I can see an issue.

Oh, one thing I did early on was bought 20 pre-poured agar plates from a place with good reviews. Once I was able to get some of those looking ok, then it helped me figure out what else I had to work on. They're expensive, like two dollars each IIRC, but were sort of like training wheels on the way to trying to get agar down.

I would recommend you either:

Buy some spore swabs from one of the recommended merchants here (sponsors, those who advertise, but they're vetted here.)
https://www.shroomery.org/sponsors.php

Or post enough posts to ask for a free one on the Marketplace:
https://www.shroomery.org/forums/postlist.php/Board/43

You can even just say "thank you" when you read something useful. That counts toward your total.

Spore syringes pretty much suck. Swabs are better. Spore prints are even better than that. They give you a lot of material to work with. It only takes a very tiny bit of one to inoculate several plates.

Welcome.

EDIT: I just looked through all your posts. I think you had it right on your very first post (copied below). You got the answer (Rhizomorphic Mycelium, which is good) in the next post from verum.

The "the icicle like growth" in your first post's plate is really good mycelium. Do you still have that plate? You could transfer that part to another plate and maybe get strong usable mycelium from it.

There might be some bacteria right above it around 2 o'clock (picture an old clock face),I can't tell if those brown dots are on the try or under on the table.

But other than that, there is a lot you could probably transfer away to and do well on that plate. The non-icicle parts that look like fluffy clouds are Tomentose mycelium, also good, but people usually think that Rhizomorphic is better. It's all good, but people get more excited about rhizo, it looks more organized and sexier.

I would take about 8 or 10 tiny transfers from that and go to 8 or 10 new plates. A few of them will probably kick ass. if any are yellow again, toss those.

By the way, that's called a Germ plate (germination plate.) If you transfer a tiny slice of that away to plates, those plates will be T1. Transfers from them will be T2. etc. By T2 stuff is usually clean enough to send wedges to grain. (Don't try to do Liquid Culture yet until you get wedges down.)

So this was a spore syringe, trying agar but this doesn't look like any of the pictures I've really seen... What's all the icicle like growth?

Thanks!

Are agar additives really necessary? Compared to straight gwa ?

Most of the time I'm just using the water from boiling rye barley wheat, any real advantages with additives like nutritional yeast?

Hi!

It was suggested to me a few pages back (adding a nutritional yeast), I've been getting some real thin translucent mycelium lately.

I've a couple of swabs that I'm considering to put to these plates (psilo cyan and a pan cyan) so time will tell. I'll also hit some of the stack with a few transfers of the see-through myc and see if it springs into life.

Not the best quality pictures, I still haven't figured out the best way to take good pics of my plates  anyway, I'm about to take the final transfers of these varieties (yeti, ghost, PE, melmak) and get them ready for grain. Anyone have recommendations about where I should take my transfers from?

The melmak cultures don't look very good and I think they're fighting some contamination, I'll end up xferring from the top one and leaving the bottom two alone

Nice Derrayld! That MVP mycelium is real easy to work with in my experience too.

I'm about to birth 2 tubs from that generation next week.  Excited to see if those fat boi phenotypes stuck.
If so those tubs will be HEAVY

Groovy germ plates, SB.

Though is that penicillin...the round spot on the left of the third pic (Alcabenzi [first] germ plate)? Damn condensation, can't tell. The right side of the long swab blob should be great to transfer though.

Does anyone grow Penicillium intentionally here? Wouldn't be bad to have for the zombie apocalypse, if it could be extracted. I've thought about it.
