Go for the 500 ml., I make uop 400 ml agar with which you can pour ~20 petris. It seems innocent. I now have 8 whcih just fit the 23-qy presto. you are going to be blasting through agar and petris. I just took delivery on 1000 celltreat petris. i am in love.

I saw those on Amazon. These are the ones I picked up for a quick fix. I'm going to invest in something like these once I get the groove of things.

I really like the idea of the 250ml's just for the simple fact that I'll probably do a bit of color coding, but I hear what you're saying regarding quantity.

Apologies for the newbie question, but are these reusable or are they a one and done deal? I'm planning on going to glass for some of the long term storage stuff, but I haven't had any experience with disposables/plastics yet.

So this is what I expect to be trich mycelium piggyback or another mold. The very leading edge you can see healthy rhizo mycelium, but everywhere else is a thin layer of fuzzy growth covering the mycelium.

Does anyone have an idea of how you end up at this point? I've sometimes been in the bad habit of not taking transfers from the very leading edges, which I suspect doesn't help the cause. But at some point I am literally grabbing this mold from other plates most of which originate as clones, and can't seem to get away from it in some cases. I'm trying to nail down where it originates if it's probably in my choice area of selection. If a plate is technically clean will grabbing from or near the center still pose a risk for contam on further plates?

I've been using a new corer tool to make circle cuts, and potentially I'm not sterilizing it red hot in between transfers, my scalpel blade gets red in 7 seconds but the tool is thicker, and I also only sterilize for 7 seconds, maybe should bump that up to 20-30.

They're disposable.  They're made of polyethylene plastic which melts lower than heat sterilization temps, they're sterilized after manufacture via gamma radiation.
Polypropylene (often referred to as 'PP5' around here - PP being its common abbreviation, 5 being its recycling code) plastic can handle heat sterilization temps.

I once moved over to metal straws for circular transfers.
You get them glowing red faster than your scalpel.
Also the angle makes them very comfortable to work with, maybe give´em a try

I'll stick to the PP5's until I do a bit more research then. Thanks for the information, everyone.
