Testing out the new flow. Letting these plates sit for 24 hours. If the filter is operating properly they should be fine right? My gut tells me that 24 hours is a long time for something to wrong when it's not supposed to.

Shouldn't take 24 hours of continuous run-time to know if your hood is working properly. I would run it for like 2-4 hours tops. I tested my hood by working in front of it. Congrats on the new toy btw

Let it run for an hour, put the lids back on and put them out of the way for a few days and then check em. Talk to aka, that dude was super skeptical of his hood lol

Want to give a quick shoutout and say 'thanks' to d3monic for his agar TEK..by far my my clearest plates yet! Well, until I forgot to turn the heater back on in my 'lab' and they condensed all to hell :mad2:

Brownbear - that ESS myc is gorgeous! Do you usually get a lot of rhizo growth with it or is that the standard?

I still work in front of it similar to working in a SAB. It feels wrong leaving plates open for long periods of time lol. I'm letting some caps print in front of it  and will put it all away when I get home from work so it'll end up being closer to 18 hours. The only reason I want to test it is because it has 3 settings. Its kind of loud and I'd prefer to run it on the lowest setting but I'm not sure at what distance the flow starts to drop off at.

Thats how I'm feeling. I have faith that it does what its supposed to but at the same time, does it really?

Any 1 cultivating Rusty White know if this is a normal sign from spore to agar?

Sorry for bad picture, hard to get this in focus right...

Rustywhyte was tomentose on my plates until T3.  I transferred a plug to my SFD slant on 12/24.  Here it is today.

I was super nervous with the hood cuz I had 40 some plates to streak :uhoh:

I accidentally did a bunch on the lowest setting and they’re fine. I’ve been running it on low for a bit while I’m getting ready to work then switching to high when I’m ready to go.

As for agar, I’m all germ plates right now.

Sort of. The apeu is a swab which I rarely seem to  get much action from, the others were prints.

I usually streak a couple lines horizontally and then drag the loop down across them so it looks like this β€œTTTT”.

I need a little help with my cultures...

Everything was proceeding normally with isolating and cleaning cultures (into T-2 to T-5 for most varieties), but my last round of transfers is showing up mostly translucent and without strong rhizo or tomentose growth on every plate.

I'm just gonna take all these translucent plates to grain.
I think I saw someone having a similar issue said it bounced back after transferring to new medium.

Not sure if you've seen this, but this might help you.

Thanks for the redirect!
It looks like he had some luck with higher nutritional content.
Will report back if I get any good results going to grain or new plates.

Didn't get any grains together for these...but I did transfer to plates with red dye.

I know D3monic said an additive in this color may have anti-fungal properties.
It is the first time I am seeing really vigorous rhizo growth from my cultures though.
Not sure if it's coincidence, but it occurred with a couple different varieties / transfer #s.

This culture is looking nice after transferring from a 6-month old petri in the fridge.

Sometimes I’d get that and then it would like spiderweb out all weird and start to stall out. Especially the more times it got transferred to the red. Other cultures looked perfectly fine. Was a gamble.

RW Plate germinated today. This is the same print that I got from D3monic a few months back and the last time I put it on agar, I wanna say it was about 5 days before there was visible germination. It hasn't even been 48 hours and that kind of concerns me except I'm not seeing features of germinating mold. I guess these just really like this agar for whatever reason. all of the swabs that I buried are going bacterial. Definitely gonna be some hot pours in the near future.

It’s kinda hard at first with rust spores cuz they mix with the agar and myc and it creates a color very similar to a lot of bacteria.

Looks good though.

A lil heavy on the spores

Lol I do that a lot too. I’ll streak a plate and then be like ...well I’ve already got everything out and open, might as well throw some more on there.

I hate when I get nothing or like two germ spots so I always lean towards too many spores. But then you end up with forests of thick myc on the ones where they all germinate.

Yea lol, I was intentionally heavy with it which I suppose could be counter productive because of the higher probability of dirty spores. Almost every time I streak plates, I only get germination where there's visible clumps.

I was also thinking "Have I even added spores from this loop?" barely see anything when I streak my plates.... I have been doing the old sprinkle technique recently, which has been working good.

Same with me.  I usually have better luck with sprinkling prints on agar that using spore syringes.  They germinate so much faster for me that way.
