I can't think of a convincing argument to avoid it other than cost. When making agar or LC you're adding everything necessary for healthy mycelium, plenty of great growers use distilled water exclusively. Eats comes to mind.

Very good to know thank you for the replies. I assume using distilled water results in clearer agar and might help with my hard water deposits. I’m curious if trace minerals found in tap water are beneficial to have in a dish? Might be insignificant though :shrug:

I’ve only ever used tap water for everything besides making syringes, and I’ve never noticed any problems, not that means there are effects I’m unaware of. I’m sure the region is a variable also.

This happened to a batch of plates I made. Too many nutrients. When I transferred back to lower nutrient plates they were fine.

Running your hood for 12 hours to prevent condensation? Seems like a waste of run-time. 1-2 hours usually does the trick for me. I'm trying to get as much life out of my HEPAs as possible.

I do like 3-4hr maybe but I pour as soon as I can physically handle it (135ish) so I have a bit of condensation at first. I’m impatient

Do you change anything for agar media used in slants vs petri dishes?

I made some plates using new agar(gel strength: >1100gr/cm2), 3% MEA (30gr ME/20gr agar/1000ml). But I think it's a bit to thick for plates, will test some 20gr/15gr with this agar next.
I've seen people use thicker, more nutritious agar in slants compared to most people here use in dishes. Does it make any difference at all for long term storage?

Funny thing is, I'm barely seeing contamination on the thick plates, on open air tests. Might suppress spore germination, but also barely bacterial, only after messing with it to get trich over it, it showed many colonies.

I find it better to just take plates poured in front of the hood, wrap em in a quick saran wrap covering and then unwrap in a SAB and leave for a few days with the arm holes covered. This eliminates 100% of condensation. Then you don't need to put as much wear on your hepa filter

But if I pour at ~115F condensation isn't really enough of a problem to even bother doing that.

Asked this in the cult general thread but I figure it's better suited to here.

I got two plates from a buddy, PE and APE but it does not appear they survived. Not contaminated but the myc on one plate is extremely fine and stopped growing, the other plate had what looks like a tissue sample that spread the tiniest amount of myc from the transfer then also stopped.

They're about two months old now.

Any hope for these two cultures? Perhaps cut a large chunk of the myc and throw it in sterile water? Perhaps make a very watery agar plate and try to rehydrate in them? I'm sure they're clean so I probably could make some watery agar in some 1/2qt jars for them to sit in and hopefully colonize. I'm not hurting for genetics but these were pre-worked and I'd love to have them in the stable based on what I've seen from their father.

I am having a rash of round contams that turn green after a couple of days -- maybe aspergillus? It's not showing in my control plates, so I'm assuming it something I'm doing wrong in the SAB during the transfer. First it was one plate, then a few and on my last round of transfers it was 6 plates.

Anyway, can I successfully transfer from a plate w/ these contams after the green has shown up? Like take a sample of the myc from the side farthest away from the contam?

Yes. Thats pretty standard practice for trying to rescue a culture... just move extra slow and be on top of your game... ive been spoiled with my hood... had a nice looking plate show some green and went to save it in the ol SAB and boy, have I gotten sloppy! Lol
