I also see these halos, but I wasn't sure if it's just a leading edge of myc expansion or a ride along bacterial contam.

Today I poured 25 plates, performed six transfers, two inoculations from MSS, and two hot pours.

Hope something good comes of it...

Its myc guys, there's nothing riding along with them.
this happens to all my plates at some point or another.

Brace yourself:  I ended up going long on this post.  Will delete and start my own thread if asked.  All insight encouraged.  Cheers!

I went from spore print to agar for the first time a couple months back and had okay success, but everything grew super slow so I never bothered cloning.  Back again using swabs, syringes, and prints--I really have nothing to compare them to in terms of viability or cleanliness or anything--I was having huge amounts of contams for a while, until I realized I was over-mistingthe SAB and handling/wrapping the plates with hands wet from alcohol--I feel like I'm noticing all my contams are around the edges of the dishes.  I've had every color of the rainbow of contam so far.

I'd like to post a few sets of pics to get some advice/troubleshooting--is there anything I can learn from the looks of these?  I've been pouring 20 plates, then doing them all with a particular variety, then taking one transfer from every single dish that looked okay, then narrowing it from there.  I think I'm taking way too many transfers, because if I'm not working with sufficient sterile procedures, then my over-compensation is just repeating the problem.  However, it's kinda worked for me, in that at least one of the plates will look okay.  I've thrown out dozens.  I'd really like to get some good ass spawn this time around.

I'm posting a couple pics of each variety--pics of the best looking ones that I'd consider transferring, and pics that maybe sum up what the whole stack looked like, plus a couple contam pics.  One thing of note is that I've been getting a lot of that clear growth/clear halo ring around the myc--I thought at first it was a contam, but now I think it's weak/new myc.  Some of the agar was crystal clear, some was chunky and dark, because I poured it super close to it cooling.  Recipe is 500 mls water, 10g LME, 9g agar.  Just made up a new batch with less LME--just felt right.

First up is B+ taken from a print with a sterile swab about 10-12 days ago.  T2

what's with this clearness?

Hogstamper: Way to keep at it. Chasing out contamination can be a very frustrating journey. Your agar recipe is standard at 10g LME per 500ml. At 5g LME per 500ml, that level seems to bring out more rhizomorphic growth. Seek healthy, fast, uniform growth before prioritizing tomentose over rhizomorphic.

I don't see anything in the plates pictured that I would want to take to grain spawn or LC, but there are a lot of good looking sections to take from. I have basically no experience in SABs and can't offer you any advice on how to work cleaner. It seems like you are noticing where your procedure is dirty and correcting it. One thing that can be very helpful is recording your sterile work and watching it. Often times people can be so fixated on one part of the process, they don't notice where they are making mistakes. I also suggest checking out D3's perfect agar transfers thread. I think a straw or punch would be good for you to use. The smaller and more uniform transfer pieces will create a standard baseline to evaluate growth from.

SS cocktail straw > punch imo these days having played with both. Straw heats up and cools off so much faster. One these days I’ll make a silicone mold of my punch and try to replicate it with acrylic. Right now I don’t have the tools or the funds though. The brass on mine is starting to soften from all the heating, will eventually need to change it out.

When I used to use a straw it was a SS for smoothies. I never got around to making a grip and damn did it get hot.

How are you cooling off your flamed equipment? Lately I've been flaming multiple scalpels and letting them cool off in the flow. It's been faster for me and I never liked dipping in a plate to cool.

Let it all cool completely and wipe it off. As trypt said, don't toss them until they're contaminated.

thanks!  I def need a bigger SAB.  I told myself I'd get a flowhood it I had some success on this run.

I mean, I was going to use them tomorrow.  Guess I'll wrap them and let them sit a bit and see what happens, and cook a new batch for tomorrow in the meantime.

I just flame them, set them in the flow just like pictured, by the time I unwrap my plate and stuff they cool enough. The brass is real thin and cools pretty quick.

Had an odd thing happen with cocktail straws today. Here is the same plate yesterday and after transfers today:

It is easiest to see around 8 o'clock. When I pushed the straw through, it dragged the growth down into the agar. I only poured my first plate a month ago, so I'm still new, though I have successfully made 30+ transfers with that straw. The transfers before and after that plate were fine, but all of my attempts on that plate were affected.

I ended up cutting a slice between two attempts with my scalpel.

Hello all, Happy Turkey day! I’m thankful for all of the wisdom I’ve gained from lurking this website for a while. I’m seriously interested in agar but I’m not too confident in my skills to make it. I have the means to inoculate agar (spores) on hand right now and I was wondering if there is any trusted source that I could buy agar dishes from online? Pre-Sterilized, pre-poured, and all of that jazz. If anyone can point me in the right direction it would be very much appreciated. Thank you!

Pouring epoxy over a plate to use it as decoration.

If you open up the plate and just pour epoxy over it. Could it work?

I wonder if the myc would do something with the agar afterwards since it would not be able to go anywhere else than into the depths of agar.

I would love to preserve a plate and use it as decoration, eg: hang it on the wall or use it as a necklace. Growth on plates are kinda snowflake like, they could even be used to decorate a christmas tree I guess.

I’ve thought about this as it would make sweet coasters. Would probably need to pour thinner agar, pour layer of acrylic over. Let harden, carefully pop disk and acrylic out and pour a layer on the bottom .

Originally I expected the myc to be disturbed to the point it’s no longer attractive but after having done some hot pours I think it’s doable. Wouldn’t a set of coasters like this look sweet?

I think it would look absolutely amazing. I hope you try it out!!

Good idea using acrylic and a thin agar layer. I've heard epoxy heats up a lot while it sets. So acrylic might be a better choice.

When I get a really cool looking culture, I definitely want to make some cool coasters

No lie, I was going to resin up some plates in a few weeks
for some Xmas decorations for friends and jewelry for my ears.

The universe is wild sometimes.

But that would be cool if someone could do it before me! My plates are never photo worthy lol will update here if i do it

I have seen pre-made agar dishes on ebay and I'm sure some of the site sponsors offer them too, but they're really not difficult to make yourself for a tiny fraction of the cost! I make "pasty plates" (no-pour plates) in 4oz SATCO tubs, usually about 20 in a batch and I can easily use them all up in a single SAB session. If I were to buy them pre-made, that would be like £30 worth of material, and that would make for quite an expensive hobby. If you can buy supplies locally, you can have agar dishes ready before pre-made plates even get mailed to you

The containers used for no-pour dishes can be reused many times and agar is easy and cheap to make yourself, so long as you have a PC (which I assume you do if you'll be preparing grain jars to drop agar into). Petri dishes that you pour after the agar has been PC'd are quite fun to make and not really any more difficult, maybe just a little more fiddly to get the pour technique right, but it's a great part of this hobby. Your confidence and skill will increase with practice, just jump in and give it a go!

Yeah man pouring plates is the easiest part of agar. You’ll be surprised just give it a shot.
