I'm gonna try it some more. What the scalpel grabbed was way small and
no geometry involed.

Although I did love geometry, gimme a couple parallel, non intersecting lines any day !        :laugh2:

Looks like .223. That is about the perfect size.


It’s funny you brought up myc stuck to the blade sometimes, I’ve got a plate right now where my torch ran out but I did another transfer anyway so I had to mark it as a possible cross between ufo and Melmak. Could be awesome if both cultures took.

i have been using a scalpel to very easily lift the puck and place on new plate.

I have been slicing them in half by depth as i pick them up.

its a 223 556 case.  cut the side out so if a plug does get stuck i can pop it out.

Funny you ask that because I tried cutting through some thick rhizo and it just gathered around the scalpel so I just laid that onto its own plate and got good growth from it

Ya, a sliver of myc. I'm just waiting for the more experienced like Mushboy,
I keep ribbing him because I like him.  Or others...

But there is a thread describing how faceplanting a wedge,
the myc crawls out from under and contam has a harder time getting out.

So there's that too...

I do that sometimes.if there is some kind of rhizo growth clawing its way over some fluffy shit, i'll just scoop it off and slap it on a new plate.

Started using the 2mm tube today... its catches on the myc a bit more... to be expected.  i can get it read hot and still hold the back end... less surface area is hot at the tip, so it transfers back slower? dunno.

faceplanting the myc has a lesser effect than fully covering visible my and contams with a bigger clean piece of agar or a thin glass slide cover. the logic is that the faceplant leaves exposes edges from the original myc which could harbor "free" contams whereas the full cover "sandwich" method covers fully and beyond and only myc can make it out from under.

Some day it's extra steps but I feel it is quicker and cleaner. I mean the more time you keep the plate open the mor chances you contam. Here is my plates!

Yeah idk why x7x like to keep it s'mores. I have the ape getting crazy looking after my third transfer it looks all rizo! I'm excited! Already have LC for golden halo and ape going.

that's cool. i had one t1 go super rhizo on me. i wish i had used the 2mm punch on those.

A couple plates up for xfers tonight. Including a few plates from the agar dye experiment. Will be taking the puck off the red plate and seeing if it recovers on a different color plate or if it totally hindered it.

So much anime!! Lol fun! And God you must do alot of agar work judging by you're plates. I'm on like T4 and still not that rizo. I'm gonna hate it if all these transfer don't fruit 😖

This is pretty much my point. Most people doing this, with a few exceptions, are the sort of people that have trouble producing clean cultures/spawn to begin with. Adding extra steps will not help.

Though there may be fewer hand motions in terms of actual cutting motions, the exposure is greater. 2 tools instead of one, hand exposure by hovering or to manipulate the plate so as not to hover, and increased surface area of transfer.

I am pretty inexperienced. Only done 100 plates so far and I work in a SAB.

I tried the steel straw technique a few times because I think it looks beautiful. But I found it to be very inconvenient.

If you don't bend the straw, your whole hand goes over the plate. Bending the straw is better, but still kinda awkward motion to stamp the plate with. And the plates I did this with got contaminated.



Also found it to be more work to switch between tools:
First flame the straw, then cool the straw in the receiving plate. Then stamp the area on the plate I want to transfer. Then flame the scalpel. Cool down scalpel in the receiving plate. And finally moving the stamped agar piece with the scalpel.

By only using scalpel there is much less movement and faster transfer. But of course doesn't look so nice. If I had a flow hood I guess it would be easier to work with a tool like that.

When you become proficient, try it again as see how you do.

That being said you should work with your media on its side so nothing ever goes over media.
