Yeah, two longer parallel lines and then you cut through to pull multiple transfers from a row basically. It's a good idea and I'm gonna try it out

Your stealing my idea! Buy a quartz tube and change the handle..... Heats up good it's clean and heat don't transfer to far with quarts..

Good read that's a pretty slick method. I always trip on how well my myc does in pf jars compared to agar plates. Definitely gonna try this.

I felt the same way when I first saw it, then I stuck a bit of a stainless steel straw on the end if my scalpel and tried it out, its surprisingly efficient and less movements than cutting wedges ime.

Its like a poke without having to sterilize a whole syringe and squirting water

I tried a star shaped one once, too many angles, wedge got mega stuck. I think that the wall thickness will cause your wedge to get compressed into the hole and stick in it. But don’t hurt to fuck around and try.

I tried this the day before i found the cocktail straws at target.

The hex tool made a mess and takes 3x as much time to cool down as the straws.

The hex shape did what it would be expected to do... it increased the potential for the tube "grabbing" the sample... a circle provides less points of friction for the puck. think of wiggling one of the hex bits out of those tools... if you torque it too much it gets stuck.

straws are the move. i have a ton of different diameters of thin, fast cooling tube. from 2mm to boba straw width.

Still only use the cocktail straw width because its precut to the perfect length and those silicone tips double up for a perfect handle. grippy, block heat and alcohol-friendly at the same time.

See I use straws but I want to find a quarts rod/straw I figure it would be cleaner and easy to heat and the heat transfer would be less try doing 30 transfer with the same straw burn fingers much

I've done thousands of succesfull transfers without ever once having to worry about that shit.

Extra steps are bad. Less is better.

I'm telling you, its less movements. One for cutting one for picking up as opposed to 3 or 4 to cut and one to lift.

Its not something I'd suggest starting with but if you know what you are doing its pretty nifty

This is pretty much my point. Most people doing this, with a few exceptions, are the sort of people that have trouble producing clean cultures/spawn to begin with. Adding extra steps will not help.

Though there may be fewer hand motions in terms of actual cutting motions, the exposure is greater. 2 tools instead of one, hand exposure by hovering or to manipulate the plate so as not to hover, and increased surface area of transfer.

3 simple cuts is all I make, they are extremely small. Transfer is approximately the size of a pin head.just enough to skewer with my scalpel. 4 tiny/quick motions in total.

Speaking of pin head size transfers.
What about just lifting a piece of myc off the agar.

I did because I fumbled one wedge and then poked the surface
lifting a thread like piece of just myc.

Not sure where that one went, lost track. I recall
it seemed to take awhile to grow.

But it didn't seem to need a wedge under it or over it.

The underlying wedge part of the transfer isn't the important part of the transfer, it's the transportation of clean mycelium that matters.

You'll notice sometimes that your scalpel will have residual mycelium on the blade when you place the wedge on a new plate, if you cut into the receiving plate to deposit the wedge, the cut in the agar will grow a satellite colony independent of the new wedge.

So long as clean mycelium makes it to your receiving plate it doesn't really matter how it gets there. The same thing happens when you transfer a wedge to grain and then give it a shake. The wedge will deposit little fragments of myc on the surface of some of the grains to form satellite colonies independent of the wedge.
