I believe the rationale is that should your culture be otherwise clean the only other reasonable place for sneaky contamination to get in would be near the lip.

As for the isolate  I don't have any experience with them so I won't comment.

If your plate is clean its not a big deal. If you're worried about the edges (I never am) just cut 1 cm shy of the edge and leave it on your plate while taking the rest.

Happy to hear that thank ya sir

Also, 3rd transfer from clone looking like an isolate, could it be?

Suuuuper limited knowledge here so take it with a grain of salt but it looks like between 1 and 5 you got some stunted growth. I'm not sure if thats from something else and this is an isolate being inhibited or of it still needs one more transfer

I think the culture is off center.

Considering I'm high and you've been at this longer I'm gonna say you're probably right, but I still think it's stunted slightly

Looking great Lenz, like a pro. Take a couple back up xfers from it and send it.  Worse case you have backups on hand.

I'm an agar noob, but I have a couple germ plates (vendor MSS drip to plate) that are looking good still a few days after transfers. I don't see any visible contamination, so I wanted some advice on what to do with them next. Thanks in advance for input.

Here is a picture of three "healthy ones" for reference:

1. Save them in case my T-1 plates go to hell (those plates are about the size of a nickel with no issues)?

2. Just let them go as far as they can and fruit in the dish on the shelf? I think I saw this mentioned in some older threads as a way to harvest clean clones.

3. Wash them down the drain and trash the plate?

4. Spawn to grain or other medium?

5. Keep taking T-1 transfers to grow out? This seems unnecessary since I have five clean looking transfers to work with already from this variety.

Toss them on a shelf and learn from them imo. Keep eye on odd growth, in a few weeks- month or so take not of the areas that went south and learn to recognize that kind of growth/ appearance

That's not a bad idea! I am planning to do fewer T-2 transfers because it seems I started with a bit too many. Do most people just pick the best candidates from each round for transferring, eliminating/saving the rest?

i am struggling with the same thing.

corumba brazil looking so good at T1... i took T2s from that one at the bottom that went super rhiso... not sure what to do with the rest. Straight to grain?

I don't think agar should be washed down the drain. It's organic waste.  It's a common misconception that garbage disposals allow you to now dump food into your sink. It still doesn't belong in the pipes.

I dump out my agar puck into the green bin, rinse out the dish, and that goes in the recycling bin if it's plastic or washed and into the oven for sterilization if it's glass.

Z-strain spore print streaked onto these plates. Usually I'll transfer from an edge with T plates. But with germ plates, should I be snagging from the middle of the whitest portion?

Some Tampanensis plates with a drop from an MS syringe. Would you guys bother with the 2nd one? Planning to transfer 11-12o'clock from that first plate in a couple hours

Took some transfer from my other T4 plates, but decided to put this grain because why not...
Came from a Z-strain 3rd flush mutant clone. Whatsup with cloning mutants? Will I get a tub of mutants?

Yeah it looks like shit, right mushboy?

Feel like I'm getting nowhere with these transfers. Maybe I just chose the wrong fruit to clone? I've heard it's best to clone 1st flush, but don't really understand why. Thought it was that those were the fastest fruiters with the best genetics, but I don't fully understand that

...Unless you're talking about the germ plate, which I thought looked good but I have very little experience.

Try taking smaller transfers. Smaller the piece the less chance of something tagging along.

My transfers are tiny. Like half the size of a grain of rice.

Or! Check out the josex poke/puck method. Highly recommend :bongload:

Skillz yo:pimp:

Sike just practice. Nothing stopping someone from playing transfer cutting on old plates.

Made 3 new plates.

imo punches are a novelty/gimmick at best, unnecessary step/complication at worst

I guess you're right on that. Next time I get bored I'm gonna cut stuff up in my sab. I'm fucking terrible at it and could use the practice. Thanks for the tips

I do two parallel lines really close together like 1-2mm then 2 more lines perpendicular to the original two.

Then keep making more perpendicular cuts to get more tiny diamonds. You can kinda see how in the pic if you follow my gibberish.
