Here is a plate that I don't get, they were roughly the same cuts in the same pattern, but it seems like one of them is just way more aggressive.  Why??

Every wedge you take from a MS plate might have hundreds of individual colonies. Each subsequent transfer might narrow that down, but your wedges are way too big to narrow it down to a rough isolate. so until you do many transfers of tiny wedges, you will still see your transfers grow out in differing rates and styles of growth.

This is an exceptional read to show how myc reacts to many, many transfers, and how number of colonies changes over transfers.

Note that none of this is necessary or even desirable for growing shroomz. Many growers here prefer MS for the robust and fast growth, as well as the faster turnaround time.

Yeah that’s the purpose of transfers. Each tiny section of myc has different genes so you want to move the fastest healthiest section to its new plate and then do it over again. That way you’re basically selectivity breeding it for speed.

I would take way smaller wedges next time, it cleans things up much quicker. Just find the myc that’s grown furthest from the wedge and take as small a piece as you can. Assuming it looks healthy.

I would prob go 12 from the top colony and 1230-2 from the bottom right colony.

The conventional advice here @w00tmycelium is grain of rice sized, but I find that can be a bit too sticky to the scalpel. I personally do squares around around 3-5mm2 big. I also usually do at least 2-3 transfers before doing any agar to grain or other innoculant.

SAB work ran smooth this time.  getting better with parafilm.  Let's see how clean my LC syringes are... dropped a couple drops of LC on each plate about to enter the world of isolation!

Next time do what I did when I upgraded my SAB: Get a tin of cheap instant coffee, dump out the coffee and clean out the tin. Use the rim of the tin and a marker to mark out where you want the holes, nice and low and just short of shoulder width apart.  Heat the tin on the stove until its red hot, opening at the top. Then take your tote you're making an SAB out of with arm holes already marked on it, and lower those markings onto the rim of the hot tin. It melts perfect circle holes that are smooth to the touch.
You'll also be able to save the solid circle piece of plastic for your little doors.

You can just use a round file to grind off some of that rough edge in the mean time. Even just an olfa knife to cut some of it away. My hole-saw bits tend to do the same thing, but it usually peals off pretty easy.

Hawaiian pes 2nd transfer from MS


Gulf Coast 1st transfer from prints (thanks to a shroomery member who sent the print for free!)


In the gulf coast you can see some circles and X's. Those are contams or maybe the chunk rolled/wouldn't come off the blade and are suspicious but either way, do i transfer asap or let the myc grow out until they get near each other.

someone explain to me why most people never mention using an inoculation loop... it seems like that would be even better to isolate very small samples rather than cutting a small piece (which would be way bigger than what gets rubbed on the loop).  I have one but have only used it to lift pieces of agar like you guys.  And in case you're unfamiliar, you can sterilized the loop with a torch like you do with a needle.

I use one often. I find the triangle head "scalpel" Paul Stamets sells to be more useful.

yes but I'm not talking about the 'lifting' of the agar piece... I'm saying why not just do a tiny rub with the loop rather than cutting out a piece agar that would undoubtedly have more strains on it than a loop-rub?  Seems like there would be much better isolation this way, but I see no one mentioning it.

Yes I often rub, scrape, and mush with the scalpel as well. I use the tip (point) to grab single rhizo's and tiny bits of myc like pins and such. You don't need to only use a scalpel for "cutting". You can xfer anything you want with nearly any implement.

If I need to do lots of transfers or streak plates the loop is my go to due to how fast you can flame sterilize it over and over. The scalpel takes time to heat up.

I also have a set of tweezers I'm quite fond of.

I do love a good loop though. That's why I'm Nichrome!
