Kinda idiot question as I used print before but now have a syringe. Do I just put a drop of solution on top of the agar or poke into it?. I could put a larger drop on and streak from it?

I have a self sterilizing inoculation loop. I made it with a vape mod and rda. I just stretched out a coil leaving one small loop. It heats red hot in a second with no flame

Drop & Streak.

Thanks for getting back to me, I’m sure it was Mold but was kind of in denial so you’ve confirmed my suspicions nice one

Luckily I streaked two plates from the same syringe and this one seems absolutely clean.

Unfortunately what always seems to happen I have to go away for a couple of months tomorrow so even if I transferred away from the plate with Mold, doubt it would have time to recover :frown:

I know it’s not ideal but those plates will be going in the fridge till I get back (worked last time ok with cubes) I’m half considering just cutting that Mold  out before storage...what have I got to loose eh?

Damm I hate my job....:frown:

I am excited, it seems like you guys are getting growth on plates in just a few days. My second round of plates are all contaminated but a few finally have growth 11 days after inoculation! I was worried my syringes were all duds. Should I bother with trying to isolate this mycelium or start over? I was using grain wash agar but switched to LME. Almost the whole plate grew this white whispy stuff I originally thought might be whispy myc due to shitty media but is not.

Are both of these large spots mycelium or just the more solid of the two?

They look like mycellium to me.  Take 3-4 transfers and find out.  Top one looks better than the bottom

Top transfers between 10 o’clock and 3 o’clock.

Avoid the mould if you can or the plates will fill with mould.

Hi mush community. I think it is about time to make my second transfer. The growth appears rather uniform on both plates. Do I look for denser areas on the leading edge or even spread? Let me know your thoughts.

Generally we look for speed and vigor. . . The faster the myc the better. . .

Well that's what I keep hearing. . .

So if speed and vigor is what I am looking for then would I want to take transfers from 7 oclock in image 1 and anything between 9-2 oclock in picture 2?

So I have a few questions. It seems like people generally have growth within a handful of days after inoculation. I had no growth for 10 days besides molds that appeared within 2 days. One thing I would like to know is if my conditions are to blame. I've moved into an old house that hasn't been staying very warm during the nights, I also keep my plates in a room that gets no direct sunlight. What temperature and light level should I be keeping them around?
Are there any tricks to maintaining proper growth temp in a shitty leaky house for the winter months?

Cold weather definitely slows down growth, but 10 days is a long time imho.

As far as heating, you should use a space heater, an infared reptile heater, ceramic heater, etc. The general rule is to heat the space though, not the grow itself.

10 days isn't long if you're working with spores, i had galindoi spores take 22days before I seen anything. Wet/soft agar is the key imo, sometimes it just takes them a couple weeks lol, as long as the plates dont contaminate then you can leave them for a while to see what they do.

Jesus, that's a long time. I guess I need to recalibrate my expectations. I'd been tossing stuff if they neglect to show growth by 14+ days.

Here is a pe plate with some form of mold its competing against, its pretty cool how the mold just goes around the mycelium.
