You may want to invest in a temp gun. You can get one on eBay for around $10 with free shipping. You can also get your media bottle at the same time. The temp of the agar while pouring is pretty important.

As long as it’s not pooling and is just a little foggy it should be okay to use. Try it out and see.

It goes into more detail about the metal cup trick I was talking about too. You can still utilize that now if you want, I think.

“how do you fix them?

use a cup with warm water and a flat bottom, put it on top of the dish for a few seconds-minutes (wont hurt if the water is warm-hot)
and boom your dishes are crystal clear again.”

Hell yeah man. Thanks for that. My backpacking cup is doing my wonders lmao. I remember that thread but from a few months ago lol.

Forgot about that. Working well! I'm going to buy a gun & media bottle from amazon as well. Want to get good at this!

How many transfers did you do to get those plates??
Also, when you make your master jars.. do you dump your whole plate? Or will you do selections and make like 10 master jars?

I have found letting the agar cool in the SAB, and placing a dome fixture with an LED bulb over my stacks of 10 to have narrowed it down to 2 plates: second one's being a negligible amount, and clearing up in a few days... I feel the ambient temperature in the SAB  (or even the room) should be higher than normal: decreasing the cintrast between the inside and outside temps of the top surface of the lids... it's why stacking works so well :shrug:

I've also seen some put cups of hot water on the top plate to eliminate condensation all together: my SAB isn't that tall lol

Do you think my plates will be okay sitting in my closet like so without drying out for a few weeks?

The hot water cup did the trick on all of them! Next time, I know what to do. It's all a learning curve:rockon:

As long as they're wrapped, should be fine... I'm considering a larger tub for SAB... I'm building a flowhood, but will likely still pour in a SAB

Why would you pour in a sab if you have a hood?  Once you start working in front of the hood your sab will become a plate housing.

Condensation isn't a bigger issue with flow? I'd love to avoid it all together...(both SAB and condensation)

transferred a single rhizomorphic rope from a clone culture in cold storage since june... digging the leap growth!

I don’t worry about condensation...pour in a stack and pour ahead of time and it’s never an issue.  You’ll rarely do any work in the sab after working in front of the hood.  At least, that’s my experience.

I don’t wrap them until I use them.  I just keep them in a gallon ziplock on the bench...you can fit 20+ no prob.  They keep for quite some time this way and never contaminate....just pull them out as needed.  And yep, I wait until they’re cool before putting them in the ziplock.

Any condensation is gone after 20 minutes of cooling in front of the hood. Mine either go back in the original sleeve or 20 in a gallon ziplock

T1 means transfer one from the original spore germination plate. This was one transfer. Sometimes you have to do more to get the culture you desire (clean & strong). Some people isolate too, I’ve yet to get that advanced.

I personally use one plate per jar for faster colonization. The faster your jars colonize the less chance contams can take a foothold. I cut the agar into four sections, cut the center out, cut around the whole outer edge, leave that in the plate, drop all four slices in my jar, and shake it up once I get all my jars done in the SAB.

People say the outer edge can be more of a vector for contams.

I've been splitting plates to 2 jars lately and not noticed a significant loss in speed... my oat prep is more consistent now, may do side by side next time: have been considering 4 jars per plate... if it's colonized to the edge, I use it :shrug:

Huh :shrug: maybe I’ll try splitting them too.

I usually just do two or three master jars per variety anyway, so I’m not to worried about spreading my agar out. I like to see which plate performes better in each jar, then I use that jar for g2g and spawn the remaining ones.

Maybe I’m doing it wrong.. there’s always more than one way to skin they say.

Glad you’re getting the oat prep down. It’s cake once you start doing it more! Got ten oat jars ready for my APE master! Going to G2g now.
