Oh no... I was saying mad should grain his plates, the 2 PE6 are 3rd gen agar pin clones, and APE is T1 from swab plate

hey this gave me an idea. i posted up some APE jars which are reportedly "a bit off." i'm wondering if plucking a few grains from some of these jars and putting them to agar might be diagnostic of something? anyone tried such a thing for troubleshooting purposes?


idle rambling, when diagnosing an engine, the mantra 'fuel air compression spark' is chanted over and and over to reinforce the fact that this is everything that's required for the engine to run. turned upside down, spawn jars require, 'culture hydration sterilization inoculation without contamination'. what else is required for spawn not to go south?

I am just getting back into this hobby after a few years and currently trying my first Spore print to agar.
The two spore prints I am working with are about 10 years old and stored in a box in the barn for that entire time.
After 5 days I am not seeing any growth on my agar plates.
Is it normal for the spores to take this long to germinate?
Could the spores be dead after 10 years ?
Should I just get a fresh spore print and start over from scratch?
Pic's are spores on Light Malt Extract agar 5 days after removal from the two old prints i had.

Iv had spores take over 3 weeks I'd give them a full 24days to germ. They may be dead idk after 10 years but they may just be slow. I also mix my agar lite if I know spores are going on it.

Glad to know it could take as long as 3 weeks.
I was getting a little worried because nothing was happening yet.
I mixed the agar at 10g 10g to 500mg water. should I have used less agar or LMA?

I do 6g agar to 500ml with spores or 9g agar to 500ml for tissue transfer. And I usually get about 8g or so LME to 500ml h2o.

Some of my current agar work.  Just noc’d up some Golden Mammoth, PES Hawaii and Corumba as well.  Look forward to working with some new strains.  Going to take some of the others I have going to slants.

It just means it is the first transfer from the spore germination plate.

Spores can take up to SIX weeks! Especially if they're old and dry... issuing soft agar helps hydrate the spores and speed things up

I poured these yesterday. All of them seem to have condensation on the front lid. Is this normal? I guess when i noc them I could just look at the back of them. Would just make it hard to see from the front.

Just wanna make sure I did okay on my agar. (First time)

Did you start pouring your plates when the agar temp reached 117 degrees? Condinsation is kinda unavoidable at least for the top three plates IME. I do a stack of ten and poor my plates at 117 degrees, pouring from the bottom up.

You can put a metal cup of warm/hot water on your plates that have condinsation. I just put it on top of my stack of ten and it gets rid of most of the condinsation. I wipe my cup with ISO first real good personally. I got a metal measuring cup set from Wally World, and use the 1 cup measuring cup for mine.

Once my plates colonize sometimes the condinsation comes back. As long as the water isn’t pooling on your agar surface you should be fine. Sometimes it goes away by itself.

Yeah man.. I don't know at which degrees I poured. I waited a little longer after it reached a temp of being able to 'handle' the bottle.

It was a bitch, because the SAB I had was very clustered because I used a fifth glass bottle for the agar.. and the bottle had to be on a angle to fit. So I kinda was just pouring dishes as best as Icould.  No specific routine.. just getting the agar in dishes was my biggest concern.

I'm going to buy a 500ml bottle next time. But I just wanted to make sure these will still be okay. I mean it doesn't looks pooling.. just a foggy front case.
