What sort of lids are you using? The grains look awfuly close to touching the lid, or maybe they are touching it, I'd be worried about that as a possible contam vector personally.

They're the standard Ball two-piece metal lids, nail punched 1/4ish hole, poly stuffed. Since inoculating these particular jars I've upgraded to plastic ones with SFDs, which is where those cultures in the plate pic will end up.

Jars look good! No need to do more than a shake, some do 2. more doesn't offer any benefit, in fact, too many shakes can stress the mycelium out as it needs to recover each time:mushroom2:

I feel like the lids are a good possibility of being your issue, hopefully the new ones work out!

Well mycelium grows fine in LC so the water won't be a problem, but it isn't ideal giving the bacteria a medium to move fast in. On the flipside it might make the mycelium speed up trying to get away from it.

I like the island tek Agar Wedge posted the other day and I'll try to pour some next time I do agar. For the most stubborn bacteria a water agar cabin seems to work best, so maybe pouring some water agar islands will be a good option as long as the island plate always sits flat :strokebeard:

That's a really good point, using water agar for the Island will help further cleaning. Just made me realize I didn't mention the laying flat part. Good looking out! Gonna go update the OP

I was just thinking about your double condiment cups cleanup, would it be possible to do something similar there, like having one of the cups being water agar :strokebeard:

Haha, yeah I can be clumsy and is probably some of the reason I like the cabin over trench, trap door, sandwich and island

Quick question my smart friends

This germ plate grew out to this point and has stalled without reaching the edges, I know with regular transfer plates that’s a bad sign but does the same apply with germination plates? Both possibilities make sense to me so QQ time

Germ plates usually have many contams riding along. And also the different myc colonies end up fighting each other.

I've noticed with germ plates the colonies can kinda sense the space and nutrients a plate has n sometimes stop growing n go into a kinda preservation mode when transfered to a new plate had em takeoff and continue growing then fill out the plate

Could be like stated above there's hitchhiking contams as that's pretty common from spore

And on another note monos tend to not fully colonize a plate though can be transfered and will grow out some more

Just saying what I've noticed from little experience.

Thanks tweeq, I know in general spores are inherently dirty and require cleanup but the second part answers my question I believe

right my transfers from this plate have grown out great and fully covered there respective plates, I guess its just not something I thought too hard on before I just assumed the different myc colonies were having mexican standoff sort of ordeal

alright alright, thanks for the reassurances

So right now I have TTBVI clones (originated from a bacterial culture) on water agar and its getting close to half dollar size and I need to make a transfer.

Its on water agar right now. Should I pour water agar for the island plate? Or should I use nutrient agar for the island to give the mycelium that makes it across a boost onces it gets there.

Im just now starting my culture cleaning journey so im obviously full of questions lol.

How much is too much?

Would going from water agar to island tek to cabin to regular plate be too many steps? Or does one keep putting it through cleaning teks until the culture is clean? Do whats necessary?

I didn't have much luck with that, microwave seems to be the business, for the brand I have.

Hi all. Does anyone know the cause of this? It kinda looks like the cap is breaking apart. It’s only affected a couple pins so far but I grabbed them out in case it’s more serious. Bacteria? Overwatering? Thanks

Here's a question I've never considered.

Let's say you've got some jars 100% colonized and consolidated. you break and Shake before going to bulk. So that you can watch recovery and make sure nothing's hiding..

Few days later.. you've got great recovery.

Is it now okay to go forward with going to bulk ? Or are you supposed to wait for it to fully colonize again.

I feel like I always just went for it at the sight of good recovery.  and never waited for full colonization after that final b&s.

But I've been away for about 9 months and can't remember.

Ps. Preferably people that do a final break and Shake to check recovery before going to bulk  answer please.

But all answers are welcome.

Looks like rosecomb mutation, have you been spraying your fruits with iso by accident :strokebeard:

It also looks like you're growing Toque genetics, if that's the case many of them are highly unstable and will throw mutants often, should be fine either way

I usually do the final break&shake check at around 95% instead of 100%, so if they recover like they should I just let it colonize everything and spawn, to answer your question, if you know everything was colonized before the break&shake test you can spawn as soon as you feel confident they are recovering like they should.
