Island clean up * 24
    #29695565 -

Hello everybody:wave::mushroom2:

This is a little trick Ive been using for some time now and wanted to share. It involves pouring thin plates then dropping the wedge being transferred onto the glass( if using plastic wait a couple seconds so you don't burn a hole in the plate), it is essentially a trench, but without the extra step. This is ideal for SAB users who pour plates as it removes a step, and therefore a potential vector. I've cleaned up over a dozen varieties this way.

Step 1: Pour your plate thin so that a section isn't covered with agar, you will want to section at least the size of a nickel for ease of wedge drop. Sometimes you may pour less than expected, that's okay too. Before moving the plate into the stack wait 5-10 seconds for the agar to set, otherwise the agar will cover your island. Alternatively, you can minimize the wait by pouring the islands on the final pours, considering the agar will cool more by then. Allow the condensation to evaporate before the drop, best limit the potential of contamination to crawl with the mycelium.

Step 2: drop the wedge on its island.

Step 3: wait for the mycelium to reach "land" and then transfer to another plate. Be sure to keep the plates level to avoid accidentally transferring contamination from the transfered wedge onto the agar.

Bacteria and some other nasties have a hard time crawling across the dry surface, while mycelium has no issue doing so which is what makes this effective. Hope this helps somebody! Please let me know if you have any questions. Thanks for reading!:mushroom2:

Seems like youd really have to let the condensation evaporate but i could see this being hella effective.
I might pour a few half plates and give this a whirly whirl.

That's a really good point, I'll update the OP with this disclaimer. Thanks Beef! Never thought of that since I usually don't get to my plates for about 12 hours after pouring.

Wait I love this, I don't even have to cut and I borderline have a couple of these plates by accident when my flask is reaching the end of it's life

Brilliant writeup  I might use the whole backup gar jar for half pours as I do have alot stuff I need to cleanup/germinate still lol

Like doing a trench with no wasted agar. I like this, will keep this in mind next time Iโ€™m cleaning up

I've wondered with trenches and cabins if there would be an upside to zero residue where the agar was removed. And this is a great way to have zero residue.

Neat. Will definitely do a couple of these when I pour next.

I've never looked into trenches and cabins because it sounds too difficult. Now this tek I've already done on accident

Except for the transfer into the desert. Easy next step!

The ocd in me wants to see like a little ring of some kind to make the desert a nice consistent shape but i cant think of a way to do it without a vector.  A rubber band would surely survive the pc but then what?  Nah this tek seems pretty solid as it is.  Im thinking ill pour a "beach" half plate.  I got this turkey tail culture whats been nothin but trouble.

Every once In awhile I get a nice shaped desert island, usually it's kind wonky though:hehehe: have had a couple peninsulas which were cool. a beach will certainly do it. Keep us posted :camping: hope it whips the turkey tail into shape :mushroom:

Didn't really have real reason to do it, but as I had a plate with kind of a suitable spot. Maybe a tad small though.

Smudged the piece around a bit there though, when trying to get it off, so probably not that good of an example, as the island probably isn't particularly dry. But good enough for my amusement.

There seems to often be that plate that doesn't quite get enough agar. May as well make good use of it! Thanks for sharing:smile::mushroom2:

Leave it to Agar Wedge to come up with a new agar tek!  I love this idea and I'm gonna give it a go next pour.
