Thanks! They came out super clear. Here's hoping they turn out clean too.

What do you guys like to do to stop your top plate from getting foggy? The warm bottles weren't enough so I'm thinking of pouring some hot water into a spare plate next time..

Yep during transfer those 4 got the green. I most likely didn't flame good enough or was sloppy n had my hand over it or something xD

those are my first transfers and first clone attempt
and the first ist a germ plate.

do you guys think there is anything usable in here? ervthing just looks like mold to me :frown:

It doesn't seem that all is mold, top plate is toast, and I don't like the trails in the condensation.

Haven't grown gyms yet so can't really tell if the mycelium looks right or not, try having a look here.

JMF and PE can be okay, but the condensation on the lid is making it hard to see unfortunately. If you set a cup of hot water on top of the plates they should clear up enough to take some pictures.

those trails might be from tilting the plate vertically since, i think it was you, advised me to tilt it so the contam is at the bottom. i know the green and the bottom one is bad but those other smaller ones are definitely not mycelium?
here are hopefully clearer pictures.

Got it, the contaminated plate might have some good mycelium and you can always try taking a new transfer aspa, but both the contaminate colonies seems to have dropped spores so don't get your hopes up.

The PE6 and JMF you could try taking new transfer to new plates. What is your plate recipe?

im happy that my agar is not getting contams anymore. i prefer having a dry setup like you recommend (i think)to me all that spraying was doing me no good. i got a permanent setup atm.
i have about 50 plates ready to go which are already like a week old.

i do like 6g each on 400ml. thought it was better to have a little softer agar since im working with old spores. (thats what i read)
maybe bit more nutes are better later on but i’ll hopefully get to that.

should i’ll be doing a transfer from that first plate then? might do it last then after first takeing those other ones. or should i let them be a little longer?

thats by the way the only plate i got to germ if that’s even myc thats why i don’t want to trash it haha (those arent gyms)

should i be doing some trenching or cabins with them? i feel like i’m in such a tough spot doing all that as a newb but it is ehat it is.
i want to be sure to have a good starting point before going to grain


those gyms are wild and just from last week so i kinda wanna get those going aswell thought i have a better shot with freshis

I forgot to say that keeping the plate upright only works with bacterial contaminations.

Nice, with a permanent setup and armhole covers, working in a dry SAB seems to work best, for me at least. Happy to hear your plates are coming out clean after pouring :rockon:

1,5% DME should be fine. Soft agar is good for spores indeed, but it will help bacteria move if you get it. I don't see any obvious bacteria in any of your plates though.

You can do a transfer from the first plate if you don't have any way of going back to that culture, but like I said, sporulating molds in a plate is though work.

Trench and cabins are for bacteria so you don't need to be doing that at this point, but practice makes perfect so while you're doing transfers of the JMF why not take an extra transfer to a cabin and get a feeling for how it works :awesomenod:

With the gyms I can't really help. I have a print and will get to it eventually, but right now I have no experience with it.

from where should i do a transfer on the JMF and PE6?

without this forum and especially your help i would have probably gave up already.

since my technique got better i will do some more spores to agar and hopefully i can get some more to actually germ. those other ones took like 2-3 weeks round about. it’s weird too that the contam doesnt show up for like a week + is that normal?

what can i do about the mold spores do something like an brf puck or something? it feels like it’s a never ending process to get going since i got all those obstacles in the way haha would be much easier and more motivating if i already had a tub going.
hope that patience will pay off in the end :smile:

I had some petris in the fridge since 2021 and took em out in 2026, clean white growth, although very stiff, when used a stainless steel scissor, inside a soft SAB (yes not recommended blablabla but better than nothing), to scrape and put some mycelium onto another plate. A week later it seems to work (germinate) so it is promising. I cannot remember how I made that recipe but... what I'm doing now: for 125 mL into 2 bottles. Potato flour 6g, sugar 2g, agar 4g, 1g beer yeast (2 varieties),  115g water. When taken out from Pressure Cooker, after 15 mins, it was too slimey and difficult to pour. Will have to adjust.

Hehe, I am probably one of few who are using DME for dry malt extract around here because I come from brewing. If I am correct and you are using LME short for light malt extract, we are in fact talking about the same thing. In the brewing world LME is short for liquid malt extract.

Blue is a go, Red is a no.





This are just my opinion when looking at your pictures, if you see something that looks clean, organized and uniform when you go to do transfers go for it.

Contaminations can germ fast or slow, but yes it's normal, if you keep getting contamination around the edges, it could be worth trying to not use the first piece of cling wrap. Just toss the first round of it before wrapping a plate, if that makes sense.

Molds are hard to deal with, so when you do get fruits, remember to clone and print so you always have a way back to a culture.

For the plate with obvious contamination, just take a tiny piece of what looks like the good stuff furthest away from the contaminations.

And you are absolutely correct, when you have more fruits then you know what to do with, all sterile work becomes easier, my sterile technique also got better when I stopped being nervous for fucking everything up, funny how that works :rofl:

Great job reviving an old plate :rockon:

I just want to add that boiling brewers yeast doesn't really add much. For the nutrients in yeast to become bioavalable for the mycelium it needs to be broken down by enzymes.

That can be achieved by autolysis, but that is pretty time consuming so I recommend just getting nutritional yeast or Fermaid O :awesomenod:
