How long do you guys Wait before using an agar to make sure it's clean?

If you’re talking about newly poured plates, I wait 5-7 days. That’s usually enough to know if they are contaminated before inoculating the plates with spores or transferring from another plate.

Bummer. I wish I wasn't such a slacker. I wanted to get on some transfers today. If only I had poured my agar a gahd damn week ago when I made it

I usually wait until they’re solid if I need them. As soon as they’re cool. Why wait? Pouring plates is pretty simple. I have confidence in you.

If this is your first time pouring plates, or you lack the confidence I have in you, then yes 7 days should show any contaminant growth.

I think you should just use them.

Hi folks-I'm a noob, barely graduated from AIO bags, but after months of lurking and reading, committed to doing The Right Thing. I'm on my third run of agar plates, and I've got a condensation issue. It's not impacting my use of the plates, but it's driving me nuts not to be able to see clearly through them.

I made a water bath using my sous vide rig, so I'm confident that I'm pouring at 47C, doing stacks of 10 plates in front of the hood, but I'm always getting what seems like a ridiculous amount of condensation. I read a previous recommendation in one of the threads to just let them sit in front of the hood overnight and it should clear up, but photo is 12H post-pour... Questions: 1/ What am I doing wrong? 2/ Should I just leave them longer?

Condensation happens man. Only way to completely prevent it would be to leave them open till they're completely cool which seems impractical to me. Putting something warm on top like a mug of hot water seems to help.  I've recently discovered that running hot tap water over the top clears it up enough for me to see inside for a bit but I use condiment cups. Might ruin a dish if water gets in the sides

Well, I am a newbie myself, but for me it helps if I put a jar with hot wáter on the first plate of each stack and let it cool with the plates.
Hope it helps.

I utilize the PP5 bead containers and typically allow them to cool overnight in either the pressure cooker or the instant pot. The following day, I simply wipe off any condensation from the lids with a paper towel in front of my ffu. While I often use them the day after running them, I also have some that I allow to sit for weeks before putting them to use.

Freshly poured before the IP run.

Quick question, Do I need to have holes in the lids of my jars if I'm not using a syringe? Just agar to grain

Yes. Without a hole the lid will seal to the jar, and even if you can get it upen without sucking contams in, the mycelium would run out of oxygen and stop growing, subsequently dying. Without a filter on the hole you will let contaminates in anyway.

That sounds terrible. Do you pc those with the lid on?

No pours have less condensation if allowed to cool completely before lidding and pcing.  With pouring, let it cool more.

Followed Beefs Wheat Tek as always and there are always burst grains. I don’t think it’s from over cooking or anything, it’s just some grains in the bag are cracked etc. is this an acceptable amount of burst grains?
