_.-=| Axman's LAGM 2025 Log |=-._
Genus: Psilocybe Cubensis
Variety 1: P.E.S. Amazonian
Variety 2: A wild find that I'm calling Tico 12-22-2024:Made a batch of
Primo Plates 45x 60mm plates with 2.5g LME and 10g agar and 500ml distilled water. I'm considering running another small run of agar to get some black agar for germ plates.
Looked through spore prints and swabs to try and decide which variety to run. Probably going to do PESA because that was a strong fruiter for me years back. Definitely going to be running my Tico Project as well, but I'm not sure if the F-1 will be dropping spores by January 1st.
12-24-2024:Decided to cook
nektar61's blagar scaled down for my needs to make up 20x 60mm plates. 250g Distilled Water, 4.68g Agar, 2.5g Potato Flakes, 3.75g Dextrose (for me this is Karo Light Corn Syrup), and 2.3g Activated Charcoal.
I do pre-pour. Probably always will until/if I get a FFU.

Merry Christmas-eve ya'll.
12-28-2024:

Got a few caps printing, and pulled some swabs today.
Prepping 10x
BRF Pucks in preparation for reviving and then transferring the old PESA spores to agar; Just in case they don't germinate on agar.

I'm pretty sure my PESA prints are from one of these two tubs I used for a giveaway circa November 2022.
My plan is to use an inoculation loop and streak plates with both PESA and Tico F-1 prints on January 1st. I'll also use one of the F-1 Tico swabs I just made. As a backup I'll also put some spores to BRF pucks.
1-1-2025:Prepped my prints, swabs, plates, and pucks, then got down and dirty.
I made an inoculation loop with some kanthal vape coil wire, I also used a normal exacto knife for cutting pieces off of my swab. I used the inoculation loop for interaction with the prints. In-between uses of a tool, I usually place it in the glass jar of 70% iso, flame it, and cool it on a sacrificial plate.

Post use pic of my Amazonian print from Oct-'22.
Since this is an older print I'm trying to revive I also put it to some BRF Pucks in case the agar doesn't germinate anything.

Everything got wrapped up, labeled, and now we wait.
Info Dump:
-{PES. Amazonian}-4x charcoal PDA plates
2x LME plates
2x BRF pucks
-{Tico}- 6x charcoal PDA plates
3x LME plates
1-2-2025:Only one plate has any action, and it's not good. One of the Amazonian plates I dropped spores directly onto from the print looks like it's growing bacteria right on top of the spores.

Bottom View
1-3-2025
The Tico LME plate that I dropped spores directly onto seems like it is germinating nicely.

The Tico LME plate that I streaked with the inoculation loop is also germinating well.
Shoutout to fresh spores.
1-4-2025:Transferred from the above plates, I tried out using a metal straw as a punch. Torched it red hot, cooled it in a sacrificial plate, then punched out a few promising places in the germination plate. Flamed my knife, cooled it, used it to transfer the punched out agar pieces.
Out of all of the plates I used the swab on, only the one I cut pieces off of the swab and jammed them into the agar germinated.

Dusty pictures
1-6-2025:For no reason other than I had them made, I put two transfers from the Tico 006 Streak plate to BRF Pucks. I'm considering making some BRF Agar to see how this variety does on that nutrition.
1-7-2025:-{Tico Update}- Tico T1 plate check in.

Several transfers are taking well, these just look the best. It looks like one of the streak plates also has germinated.
Still no movement on any of the PESA plates.
1-9-2025:Nothing on my Amazonian plates or pucks; I had plates out so I decided to use some sterilized distilled water and wet some swabs, I then streaked the print, streaked the plate, and broke off the tip onto the plate (x2).

1-11-2025:
Made 10x Rye jars using
Bod's grain prep.
Cleaned and prepped 40x petris for tomorrow morning to cook as soon as the grain jars are cooled overnight.
Kinda just went for it but I did a 1.5% ratio of 500ml distilled water, 10g agar agar, and 7.5g BRF. We will see, it doesn't look like it will be super clear, but I'm interested to see if the Tico mycelium likes it as much as it likes BRF pucks.
1-12-2025:-{Tico Update}-
Took transfers from each of the T1 plates, one to Charcoal agar and one to BRF agar each.
From each of the T1 plates I cut wedges and inoculated 2 jars of rye each.
From BRF pucks I inoculated with agar on 1-6, I scraped some of the growing mycelium up and dropped it into a jar.
Each puck got their own jar.

So today:
Plate 026 -> Transferred to 1x BRF plate, 1x Charcoal plate, and Inoculated 2x Rye Grain
Plate 028 -> Transferred to 1x BRF plate, 1x Charcoal plate, and Inoculated 2x Rye Grain
Puck 042 -> Inoculated 1x Rye Grain Jar
Puck 043 -> Inoculated 1x Rye Grain Jar
1-13-25:I did not notice how many burst grains I had when I cooked this set two days ago but here we are. I'm just gonna let them run out.

Agar and grain are getting acquainted in every jar it seems.
1-14-25:-{Tico Update}- All of the Tico transfers to grain are growing well.
-{Amazonian Update}-
I'm seriously considering switching to a different Amazonian spore print.
Could I have made another swab? Yes. Did I just yoink that one off the plate and jam it into a brf puck? also yes.
1-15-25:-{Tico Update}-The same wedge two days later.
1-19-25:-{Amazonian Update}-

Grabbed a different Amazonian print, wet 4x swabs with sterilized distilled water, rolled them in the print, then broke them off and jammed into each puck. I also took the entire print and made a spore syringe with it. I used that syringe to also put some drops on the opposing side of each BRF puck.

I'm not convinced, but this might be some white fluff growing on this contaminated plate/swab.
-{Tico Update}-
Tico Rye jars are doing well. I did a break and shake on this jar after the picture was taken.
1-21-25:-{Tico Update}-
Tico Jar 2 days post break and shake.
-{Amazonian Update}-

Forced Hydration

Not sure if friend or foe, but I did my best to take some transfers of it to 4x plates.
1-26-25:-{Tico Update}-

Tico, both were spawned the same day, just one was shook.
-{Amazonian Update}-
Many terrible things on this plate, but the transfers I took were much less contaminated.

Transferred from above plate, then today I took two more transfers hopefully to clean it up.

Two of these pucks have contamination, two are clean. Went to agar with the clean ones.
I also put the force hydrated syringe to agar.
1-29-25:-{Tico Update}-
Tico T3 looking beautiful to me.
One of my grain jars is basically fully colonized, I'm going to let it go until tomorrow night then spawn it.
I hydrated some coco coir last night using
BOD's Bucket TEK.
-{Amazonian Update}-
Amazonian T1 off of the second print's Swab I germinated on a BRF puck is looking like I might have some workable myc.

T2 from that terribly contaminated swab plate. Looks like it might be clean, I'm going to let it grow out and see if I need more transfers to get it solid.
It looks like I'll be able to save the Amazonian after all
1-30-25:-{Tico Update}-Today I spawned this jar. It's possibly a touch baccy but it smelled fine and shook up fairly easy with no hard clumps or anything like that.

I like to put the jar label on my tub as well as a date for when it was spawned.
I went with just coco coir, mixed 1 myco quart to 1.5 coco coir quart, then compressed that with a metal concrete finishing trowel, and left a 1/2 quarts worth of coir for a pseudo casing, compressed the pseudo casing on top then did a heavy mist. So a 1:2 ratio.
2-1-25:I decided to attempt a cross between AA+ and a Tico F-1 Fruit.
I took a swab, swabbed the AA+, then Tico, then AA+, and then back to Tico
Did that for two different fruits of each and put them to LME agar.

I've never tried anything like this before so here's to new things.
2-4-25:-{Tico Update}-

The tub spawned on 1-30 is coming along nicely.
-{Amazonian Update}-
Hydrating the swabs was definitely the move.

I'll be taking transfers from that plate soon.
I threw some sketchy transfers to grain while a little tipsy the other night.
-{Tico x AA+ Update}-Three out of the four plates are showing signs of germination.
2-11-25:-{Tico Update}-
Lost one to trich.

I realized today that I can use the sealing bar on my vacuum sealer to gusset the edges of ziplocks to make them form nice cube edges for Fahtloc bags.

Another Tico jar was spawned.
-{Tico x AA+ Update}-
On 2-9 I transferred from the bottom left plate, but they all have good germination. A few spots have myc leaping upward into the air.
2-23-25:-{Tico Update}-
First tub to pin, some promising clusters in here.
I also inoculated a few jars.
-{Amazonian Update}-
Taking their sweet time, I also inoculated two more jars.
-{Tico x AA+ Update}-
I inoculated 6 jars with 2x T1 plates, and 4x germ plates.
3-2-25:-{Tico Update}-
Only F2 tub I have with fruit right now. Real leggy this go around.
3x F2 fahtlocs awaiting pins.
-{Amazonian Update}-
-{Tico x AA+ Update}-All 6 of the inoculated jars are growing well.
5-11-25:So I took a long vacation and generally got overwhelmed with some major life things, but I'm back.
-{Tico Update}-
The jar was left so far past consolidation that it grew invitro.

An overgrown tub.
I took spores from the F2, germinated them and transferred, here is F3 on agar now.
-{Amazonian Update}-

Although the green scourge took this tub I managed to clone one of them to save the genetics.
-{Tico x AA+ Update}-I have several tubs going, no pins yet.
