Not all bacteria contam is noticeable in grain(to untrained eyes) at first.

But you'll no doubt see it with agar.


So right after (& before,  if you're doin a bunch of jars) you 'noc up your grain, take it to plate for validation that your grain is (probably) clean. And takes the guess work /worry out of the gamegrain

if ur certain the lc is contaminated, you will certainly contaminate your grain. however if ur uncertain, u can at least be certain of the uncertainty wether ur grain will be contaminated. make sense?

It's possible, it's also possible to see contaminated LC. 

Not always but after enough experience and paying attention whist keeping all the variables constant, you will recognize bacteria lc just like it was on agar.

ime, a cloudy lc is a nasty lc. just toss it. sediment may also give a cloudy appearance but, mycelium will eat it up and the liquid becomes crystal clear when its a healthy lc. just for example.

If this was in the Cultivation confessional, I would probably also confess a bit in line with that. But I don't always, sometimes I do test it on agar.

What I dislike about LC testing on agar, is that even the tiniest drop seems go all all round and is not staying centered in the middle.

Here comes the quick question: can that 'going all around' be circumvented, f.e. by making the agar dryer? How?

ive tested lc on agar once and used a swab which worked pretty well tbh. myc stuck to it and didnt leave too much moisture on the plate. im just paranoid im gnna contam my lc during the testing phase hah! i like to open my lc once, and thats to pour into grain. if i have a bit left over, i toss it. not saying this the safest way but i try to get my agar suuuper clean so once its in lc, i dnt worry too much anyways

Maybe after a while but IME harder agar will allow it to roll all over just as easily.


But having harder agar helps keep bacteria in place... and most won't be able to survive  3% agar... not saying to go that high but 2.2-2.5 % will do wonders

Avoiding the need to draw it out with a syringe. Ive got a fucky thumb on my dominant hand, so operating a syringe one handed is tough, and I feel like if I contaminated the LC, it will be while doing that before it's the culture or transferring the wedge to the LC, gnomesayin?

I was just hoping that a contaminated jar from bad LC would be very obviously contaminated, that's all. I may just figure something else out. Maybe sterilize an empty HG, pour the slightest bit in it in the flow, and then use a loop to put on agar.

Bc my mouth doesn't suck that hard. Ive tried already.

I considered using a loop, but I dont trust putting a tool in the jar, and pouring some out to grab with the loop feels like overhandling. I'll figure something out...

That makes me feel better, bc I dont care for the idea much myself. Could you recommend a low risk method of getting it to agar? Someone mentioned a swab - I think I could get behind the idea of just pouring a bit on a swab. Maybe just do it over a paper towel or something to catch the mess and then apply the swab to agar?

Pour a little bit into a sterilized short jar (4 oz or wide-mouth half-pint), or another sterile container like a petri dish. Then you can use a swab or an inoculation loop to pick a bit up and streak a plate. (Edit: Looks like you already had that idea. But it's the right one IMO. :smile:)

Remember to shake before pouring so you get plenty of mycelium in the little pour.

My first time making a LC from a Clean Agar Plate and transfer it with a Syringe Nedle and shoot it too the injection port after 4 Day's the Nats growing very well inside..

What you think?
