Coir and vermiculite do not need to be sterilized or pasteurized. Using hot water may help expand the coir and improve water absorption, though. Gypsum is not necessary and doesn't require heat either, but it may or may not provide some benefits, so no big harm using it if you already have it (environmental impact aside).

Nutritious substrates (manure, straw, hwfp) and casings (peat, etc) do benefit from pasteurization (or sterilization) to give the mushroom mycelium a big head start in colonization before other microorganisms can take hold. I say sterilization because many here do have plenty of success with sterilized casings and substrates. It has been argued that the benefit of pasteurization over sterilization is that it doesn't kill off beneficial microorganisms. However others argue that this does not apply to active mushrooms and is a hold over from gourmet (button mushroom?) farmers that doesn't really apply to active species.

All this to say, you are good either way, friend.

Have any of you done any sequencing?

I'm trying to grow from a wild cube and I was wondering if I could use some spores or healthy mycelium in order to get it properly genetically sequenced. Just for my own knowledge and curiosity of course.

Skip the LC and go direct agar to grain.  If you're not moving volume then there's no point to LC, it's just another contamination vector.

how long do spore prints last when stored properly? what about spore syringes? and what's the best way to store both of these?

It is my understanding that PCing your substrate gives other contamination an edge over just pasturizing it. And I personally haven't had much contamination issues using bucket tek substrate weeks after it's been sitting in my cooler.

I can mix 2000g of substrate by hand within an hour in my cooler, it still is hot af though. I personally would still probably run the sub you made.

Or just make more if you have more coir.



Yea but I made so little coir that it only stayed hot for
like 10 minutes. Isn't it considered pasteurized when it's hot for a longer period of time? Thats why I was thinking of giving it a steam bath since it cooled down way too quick

Is pouring boiling water over substrate even necessary? I think I read some people just pour regular tap water onto their coco coir/cvg without heating it up now

Are these Mycogone because they are blobs or are they Mycogone because they are reddish colored? Is the reddish color Mycogone or is it "mushroom urine" or whatever? That's where I'm having a hard time.

We use boiling water because its said that cooked coir seems to work better.and the hot water cooks it some if you put it in  an insulated container and maintain temperature for a while. . You can tell by its color. Cooked coir comes out nice and dark. Otherwise if not maintaining the temp hot water helps it expand and break apart easier for the mix.. other than that it's really not necessary

So if I have a sub that shrunk a lot after it’s first flush and I’m going into my third flush and it’s just kind of shriveling. And I have a couple of big tubs like that.
How offensive is it to just mix the two tubs together, put them back at field capacity and then compact ir back down as if it’s a brand new tub?

It may not have enough energy left in to fully recover & fruit substantially.  I just soak those fkers for like a full day. And try to squeeze out another on.

But hell if you're bored give it ago and let us know.

this would be something that would happen after maybe the third flush so it’s not terribly important that it does an amazing fourth flush. But maybe it would have a really good fifth flush.

I'm more open to it being blobs after everyone's input.

I guess the real test would be rip one of those lumps off and cut it in half.  See if it bruises.
