on old (2-3 mth) plates that don't look good, but haven't gone moldy or show metabolites, what is likely happening? bacteria of one kind and another and so there's kind of a stalemate and myc stays alive but unhealthy? lots of things are just not worth trying to clean up, right? (trying to be more ruthless about throwing stuff out) This is pan cyan MiB

I missed all the responses to my pic of the plate that I lost the nice growth on. Sorry about that. The plate was part of a series of transfers I did on 10/14, many of which ended up with contam issues. This one in particular was Golden Teacher, the parent dish having come from a BRF jar which itself was inoculated with a plin.

Suppose that 11 oclock sector is my best bet for a transfer?

Quick agar question (I know, wrong thread):

Can a change in weather make plates (holy grails) form new condensation?

I noticed it the other day after unwrapping some new ones, which were pretty condense-y already, but others that I had from before got new condensation. Not all of them have cultures. Kinda sucks since I needed those new ones yesterday. Thanks for any input on this!

I'd say your question qualifies as "Anything and All things agar"
Yes, temperature variance can absolutely cause condensation to appear. But condensation isn't that big of a deal, just store your dishes upside down

I hadn't thought about storing them upside down, thanks for the tip. And my only concern with it is visibility, especially when there are water droplets around the sides as well. Time to flip the plates!

I'm thinking you should rock that thing around the clock.  Lot's of potential..

Listen amigo - If you think I'm willing to make 3 transfers off this motherfucka, you would be absolutely right. Thank you for the input!

That was my initial intent, but the bare spots between the strong sectors made my spidey sense tingle. I did take this from a germination puck, so maybe it is clean...idk. Still learning to read plates. If there's no contamination here, then hell yeah - diversity would be optimal here for sure.

Don't need to wrap HG plates. I've had a pour plate be just fine for 2 years with no infection. They're solid.

When they fail is when there's moisture pooling on the side, contams can swim up the water where they find...air swimming?...up the threads impossible.

I think you have gotten to a point where you *feel* where you should transfer from.

Assuming the bottom is tinted blue due to something behind the plate.

I would take a new transfer to keep the culture going and send the rest

Yeah, that blue isnt on the plate - everything looks fine to me, though I will say that I didnt know the segregation between sectors could be that pronounced, but that definitely adds another piece to the puzzle. Appreciate you guys!

What is the transfer?
Plin, grain, agar?

The source of this plate? I germinated spores on a brf puck and this is the sample I took from that. I then transferred the 11 oclock area to agar earlier today.

Ok I was wondering why it isn't growing out uniformly which made me think it was a plin or grain maybe.

Looking good though

:congrats:
Way to go. Upon examining it again tonight,  I was thinking you were right on focusing on 11. 11 looks 👌 Looks like the longest, thickest, most rhizo and the fastest growing.

I guess I didn't mean "unwrap", I meant that I had just taken them out of the jars that were in the PC.
