I pour my plates, these ones I did when I had the flu and the agar was rather hot at the time of pouring.

It's still a bit cold here and I think the temperature swings in the room must be contributing to condensation also.

My latest batch of plates were beautifully clear for days, then today I noticed they had formed condensation :facepalm3:

There is a lot going on in that plate, which makes sense from your story. I’d recommend either starting back from spore or taking a tiny transfer from the least fucked but of mycelium. But the latter will be much harder for a new cultivator than just starting back from spore

Is it fucked because I left it out so long, or are those metabolites a sign of contam? Which part would be best to sample from? I can do both, take a sample and start again from spore, as I just made fresh plates and I have extras, so I may as well.

If you've got the plates might as well get your agar practice on.

And at 0:10seconds I'd start with just past 6o'clock between 6:35.
Take small samples to their own plates...

I concur that subculturing from that will be a shit shot of bacteria or yeast under the mycelium. the idea is to identify healthy, discrete colonies with agar versus ones that overlap with bacteria or random fungi as soon it looks promising. personally I'd trash it and use a more less inoculum next time.

Hey btw, syringes arent good for inoculating agar imo. Use them on pf tek cakes. Use spore prints for agar.

Syringe to agar can work just fine, though maybe I've been fortunate with clean syringes. I usually use a sterile container (e.g. 4 oz jar, these days I'd probably use a sterile cryovial) to squirt a little into after shaking the syringe and flaming the needle. Dip a sterile inoculation loop and streak a plate or two. I just do a Z pattern, all at once (I rarely do proper serial dilution).

BRF pucks are also great for germinating spores in any form. Just make the PF recipe without the dry verm layer (and with unmodified or filtered lids, not unfiltered PF lids). Transfer to agar with the tool of your choice when it's grown out a bit, and/or wait for pins or whole fruits to clone.

I had to watch a few videos to understand what was meant by leading edge and how to decide where to section from, and now I understand why it’s fucked. I decided to start over this morning from the MSS and keep a better eye on it this time. When should I look at it or expect it to be ready to take a sample from?

Regarding my crazy agar plate, just so I can learn from it, is that amber droplet likely contam or just metabolites?

Spores germinate on their own time so you kinda just have to check it once a day or two.


That's a streak plate that's just about ready to take transfers from.


A more aged example but with plenty of places to choose from still.

Amber droplet is metabolites, which on a plate usually indicates the culture is fighting something (probably bacterial).

I have been messing around with my agar ingredients as of late and doing a lot of transfers. This particular plate contains agar, sawdust, rye dust, malt extract, and a small pinch of gypsum.

I noticed the mycelium is a bit light on growth in the middle and normal on the outside as shown in the photos below.




My question is, is there bacteria effecting the growth causing the odd look or is this normal and I am over reacting? Or, could this caused by my odd agar concoction?

More importantly, you wouldn't use the majority of the nutrients.

What he's trying to say is you over nutrient that agar for zero value, hell it probably slowed the growth down.

Spores dropped 10 days ago. I am not sure because they all look so fluffy, but I can not belive they are all bad.
