Syringe Needle as Transfer Tool * 12
    #27959405 - 09/20/22 06:06 PM (1 year, 4 months ago)

Nothing mind blowing special and for sure nothing new. Just want to promote the use of syringe needles as transfer tool and maybe someone will enjoy this.

For the last 2+ years I am using a syringe needle for transfers and I'm loving it. All I do is grab a 20G needle and stick it onto a 4 inch nail which has the right diameter
for the needle to fit and stay on. It lasts for many many sessions.

The shape and small mass have some advantages compared to a scalpel.

  • the needle heats up much quicker than a scalpel (useful when using an alcohol lamp)

  • cooling the needle before taking a transfer is not necessary due to the low thermal mass

  • the fine tip allows to take very small samples

  • better visibility of what you are doing

The fast heating and cooling is the main thing I like about this and a 20G needle is thin enough to allow for that while remaining sufficiently sturdy. Bigger needles
will of course work as well but might require cooling before taking the transfer, especially when taking tiny samples. Haven't really tested this out yet though.

I use this in two ways. The fast way is to poke the needle a little bit into the agar and lift it up. That way a chunk of agar gets "ripped" out of the surface.Only thing to
look out for here is to not push the needle through the sample or it gets difficult to get the transfer off and onto the receiving plate. It's fast and works great for
expanding clean cultures to more plates.

If I need to make more precise transfers in order to clean up a culture I use the needle to cut a tiny triangle which I then lift out with the tip.

Thanks for sharing! :mushroom2:

Would scraping agar's surface for mycelium work(instead of cutting a triangle into agar)?

yes, that should work. The only thing is that the needle might still be too hot for just bare mycelium and I think that a transfer with a little bit of agar recovers faster though. Easy enough to run a few tests.

I've been using exacto's tip in the same way. On destination plate, I just cut through agar to dislocate the agar piece from tip. Sometimes agar gets stuck to exacto's tip.

How do you transfer on destination plate? Do you poke again or cut through?

On the receiving plate I'm doing the the same movement I made to lift the sample out but in reverse.

Oh shit that's cool, I really hate fumbling with wedges and they are usually a PITA to get off the scalpel blade. Much smaller transfer is the way to go IMO. My best transfers seem to be smaller ones every time.

I've been using this for a while and it's the perfect tool for transfers. Thanks again Baba!

As a handle I use an exacto. Just remove the hub(plastic part) from needle and insert it into exacto. It allows to flame the handle.

Great to hear you're liking it.

Using an exacto handle sounds like the way to go, will get me one soon.

Love having needles on hand, I use a pair of pliers to bend 3-4mm of the tip 90* so I can use the elbow as an inoculation 'loop' and also have a bit of a hook to snag grains from a clean grain-master if I'm going G2A.

45 degrees and you have a cap handler. PF never touched the caps when making syringes.

The Psilocybe Fanaticus spore print to solution device. If you bought a syringe from PF it was made using this device.

Cross post from new tools thread…My hand is too tremulous to do any small work like cutting a little piece of agar. But I can still stab stuff accurately.
16 gauge needle, small paper clip=1.8 mm biopsy. I heat it up with IR or flame, cool it in clean agar then transfer.

I have since ground the bezel down to create a uniformly round tip.

Transfers are pretty:

Nice, if you haven't seen this already, V.L has build a V.L micro-agar transfer tool out of a syringe.

looking good! Still haven't got around buying an exacto handle

Like the cap handler idea as well, seems better than the tooth picks I use.

Jealous of the cap config on yours Tiamo! Added this little guy to my arsenal and just put some time in with it. Definitely different but I enjoyed it, thanks BY

I cut off a small bit of the sides of the plastic on the needle base. Then I had some room to literally bash it in with a hammer. Also used some RTV silicone to keep it in place and fill the base of the needle. Worked quite well.
