Ah, ok….I misunderstood.  And good, as that wouldn’t make sense to me.  I’d just toss the problematic cultures and go back to spore unless those are super dope killer genetics you have to have.

I am just about to that point…there’s a few that immdef gonna water agar before just starting over but imndef getting to the point of just tossing and starting over..a few I don’t have generics for anymore except what I have going but I’ve found the marketplace tk be very giving and I’ve been paying it forward a lot lately for good karma too so I’m sure I can replace

Love when you don't look at a plate for a couple of days and all of a sudden find a yeast contamination.

The lids for these containers are press fit and some don't fit very well and I'm pretty sure it lifted a little when I handled it the other day. Micropose unfortunately won't ship the PP petri dishes to my location so I just said fuck it and went with the glass ones instead. Looking forward to actually sharing some decent agar pics with you all

I've got a clone plate I need to transfer from but it has grown mould overnight, albeit far away from the healthy mycelium. I only have a FH & I'm not loving the idea of doing the transfer in front of it for obvious reasons.

Do I need to make a SAB for these scenarios?

You could do an open air transfer

Look up "bunsen burner sterile field"

Open air transfer from contaminated plate using this method.

Thx for the suggestion

Found some funny dialogue between Josex & Bod lol

Sounds & looks like it's worth a try but I don't have a bunsen burner to hand so I suppose I'll just repurpose one of my monos with broken clips

:wtfsonic:

What a strange colour for MEA. Did I mess it up by forgetting to shake it after adding the dry ingredients before PCing?

That's some old school stuff. I've got a (professional) microbiologist friend and this is all she uses in a commercial setting.

Doesn't look that weird to me? I've had bottles of agar look nearly identical & work fine. I have never forgotten to homogenise my MEA though..

Is the texture normal when you swirl it?

It's solid, so I can't swirl it

Oh I've got one of those :stoner:

You just leave it blasting above your plate & do the work yeah? Will try it out today. Cheers.

I reckon heat it back up in boiling water give it a good mix & pour away. Colour's fine to me.

I prefer to think of it as proven and time tested instead of old school

My first two clone attempts. Also, T3s from the T2s I put to bulk and got the clones from.

Some bacteria close to the clone meat but will be transferring these to another plate and then hopefully to grain. Thanks to all the great info and people on this site!

When choosing cultures to transfer do I want the densest growth or the widest, most area covering growth? The 2cm dome of thick fluffy mycelium or the 5cm thin mycelium?

Fastest growth, so furthest edge from inoculation point.

Just dropping in to say,

If you’re thinking about microwaving your media bottle to melt the leftover Agar and then rinse it off with hot water..

Don’t.

My media bottle just shattered in my hand:banghead:
