Give these a chance to grow and share results. I know I know i know the science behind it. If I cook my agar too long it will not grow anything, and that is just a bare, naked-assed fact. I dropped my cook time and it harbored life. I don’t know why, I’m not making any claims, I’m just putting forward a solution that worked for me. Doesn’t mean it’ll work for you.

Even water agar grows myc. There’s no straightforward reason why a handful of people, new or otherwise, get dead plates.

Being new doesn’t mean anything. I’m doing everything identically to when I first started except cook time 🤷♂️

I use munchausen's agar recipe to the tea and never had issues germinating even the most stubborn of spores. It is weird that not even bacteria is growing...

The only important thing is this:

Your PC chemically can’t caramelize the starch in your media. It doesn’t get hot enough to cause problems, so cook time shouldn’t matter at all. You physically can’t “overcook” your solution, so it shouldn’t matter if it’s in there longer than 30 minutes.

But I only have success between 20-30 minutes. That’s not a disputable issue. I don’t know WHY, but that is the truth. Like 5+ batches of dead plates with known, established recipes say so. Besides that, the recipes aren’t ever a problem. Agar + nutes + water = working plates. Everyone agrees on that. So why are we getting barren plates?

I get plates full of LIFE at 15-18psi.

Had. I was cooking for 90 minutes at 15psi and they would either grow nothing or myc would stall out at a dime sized diameter. Even contam, if it germinated, would stay pin prick sized.

Dropped it to 20-30 minutes and never had another problem.

Wonder if sterilizing for too long denatures any of the nutrients? I know this is kind of a sore topic, but im genuinely interested. Have you ever gone back to pressure cooking them for longer and seen if its still not germinating any life, or tried different nutrients?

I said I was going to do a run following my old protocol last time it got brought up, but then I forgot lmao. I should. Worst case scenario the agar works and then we have no fucking idea what causes this phenomenon or how to fix it.

dude investigate these two things you will get the answer.

Overheating:
Prolonged exposure to high temperatures, such as 45 minutes of pressure cooking, can potentially denature or destroy essential nutrients, growth factors, or other components present in the agar medium. This can create an unfavorable environment for mycelial growth and inhibit its development.

Nutrient Loss:
Extended pressure cooking can lead to excessive evaporation of water from the agar medium, resulting in nutrient loss and altered agar composition. Lack of adequate nutrients can hinder mycelial growth and development.


also better follow that agar cook time chart and temperatures.

i have used only PDA agar

The melting point of starch, which is a major component of potatoes, is typically around 200 to 212 degrees Celsius (392 to 414 degrees Fahrenheit).

if you over cook PDA nothing will grow on it, potatoes starch don't need to melt to lose nutrient they become useless because nutrients are loss. turns out starch is not the only thing mycelium need, some sugars are gone in potatoes as well to compensate for sugar loss we add dextrose.

The melting point of dextrose is approximately 146 to 150 degrees Celsius (295 to 302 degrees Fahrenheit).

you don't need to hit this temperatures holding 115c to 120c or 239 248 for long period of time will destroy it

the one of the reason autoclave and pressure cook operate on time and charts exists its very reason NOT OT OVER COOK IT

yes

dude i got 13 psi and i don't go over 25 minutes. Borosilicate Glass melting point is  820 to 860 degrees Celsius (1508 to 1580 degrees Fahrenheit) but that does not mean you can stick glass petri-dish in oven.

same goes for pressure cooker, you have to follow the time rule.:shrug:

The only folks with this dead plate problem are noobs. Not one TC has ever experienced this issue, so far as I'm aware.

Hope it grows mold, I really do.

The whole point of it all is that I followed a recipe exactly, high quality ingredients + distilled water, cooked it for 40 minutes, poured, etc. Before wasting time doing transfers to non-viable agar dishes, I tested one by exposing it to very dirty air for 30 seconds, 4 days later I did it again and added a dab of spit (I know), waited another 4 days still nothing. Now that plate is the same that the mold was added to. If I can't purposefully contam a plate I can't use it to determine my sterile technique or the quality of my cultures, even if it DID decide to actually grow out some mycelium. This is my concern, and ultimately the issue i am attempting to troubleshoot

I have always sterilized agar for 45 mins at around 18psi. The sterilization charts for agar can exceed 45 mins @ 15psi, don't be ridiculous.

agar pressure cook time is 25 minutes you can not go beyond that or it will be over cooked agar.
same goes for grains 90 minutes or 120 minutes go beyond that you will end up with burnt grain.

i guess stipe-n cap has that chart thing with agar times on it.
