I don't think anyone is messing with you mate.
I just did these plates just a couple days ago using Stipes exact method.

What you're seeing is the slight condensate on the lid through the agar itself.

I know yall are truthing I just can't see why the massive difference in results, mine all look like this with stipe's method

Actually that loop wouldn't be too bad as a booger hook.

Yo Tri-Polar do you have any way to cook it at less than 15psi? I used to get it pretty clear on my old PC that only went to 12-13 psi (allegedly, no gauge), I used to cook it for 45mins. I can do a test run tomorrow with it and see if I still get it as clear.

I'm liking the way the 1% and the .2% looked especially when I poured them, gonna see how they perform against the 2% and if its comparable then boom, I'll gladly use less LME :shrug:

I imagine you guy's are using some kind of shitty malt. I vaguely remember this sort of shit happening to people using diastatic malt.

I use this. I never have iasues, ever, not even once.



Rule of thunb: if you've tried every available proven technique to no avail, change your materials.

Dry malt extract @ 2% by weight
Agar @ 2%
Distilled water
45 mins @ 15psi

No problems. If you cannot make that system work, it's your malt. The agar shouldn't be relevant.

If the 1% or .2% don't suit my fancy ill buy that brand, I have tried multiple brands but yours looks fancier than what I've tried so we shall see  :shrug:

See I read a tek a while back talking about water agar for cleaning up cultures and how fast the myc spreads out looking for nutrients and I've been fucked up ever since

High nutes = slow lazy myc, low nutes = fast hungry myc, too low nutes(<2Ωͺ) = slow starving myc?  Is that correct? I'm really slow :smile:

2% nutient ratio is moderate, consider it to be the line which separates low and high ratios. Above 2 ia high, below is low. Water agar is used to clean particularly filthy cultures for that reason, the lack of nutrients slows the growth of both mycelium and contaminant while allowing the target mycelium to outpace contamination due to it's exploratory impulse to locate resources. That exploratory phase is faster than contamination but slower than normal growth.

Water agar is used to slow the growth of individual hypha when isolating monosporic isolates for breeding/crossing.

Unless you're attempting to outpace some mold, aggressive bacterial infection, or produce monospore iaolates, I'd maintain a 2% ratio for standard culture work.

There shouldnt be any foam to scoop, we're not brewing beer. I don't know what spray malt is but I'd personally stay away from it if you're having issues ( Correction: it appears to be standard DME). The same applies for diastatic malt, or anything else that causes proteins to crash. Get yourselves some proper light dry malt extract, briess malt is often used.

As for the foam im referring to when it goes to boil over when doing a pre-boil on stove or in the microwave. Mine gets quite the head, lol probably another testament to my DME quality

Actually I have a bag of briess in my cupboard, its what I used first but had the same issue :\ ill be trying the brand you've linked

Though I may do ONE more test when I get home just to make absolutely sure the briess aint cutting it for me. We shall see

Oh I know, I read the links a lot and often, had just seen another TC mention its what cleared up their agar (honestly thinking back they mightve been talking about DME LC) so just wanted to give it a shot, if it fixed the boogers I wouldn't mind the extra step

Yeah, so LC is different. With Liquid culture broth I use .1-.2% for the sake of clarity. The culture doesn't grow as quickly, but faster than it would on agar due to the benefit of stirring to break up the colony which accelerates growth. This can't be done with low nutrient agar.

I sacrifice a bit of speed in order to gain the ability to quickly identify contamination due to the clarity of the broth. Although slightly slower than say 2-4% broth, it's faster in the end because I won't need to confirm on agar prior to use.
