I think the cost of diminishing returns with the slurry is medium/medium-high. The difference may be observable, but I don't think it will be significant enough to warrant a second attempt. Let us know what you find out though if you do attempt for sure, be interested to know.

I just did 8 traditional transfers today, but still may give the slurry idea a go. If I do, I'll absolutely report my findings back here good or bad. Worst case scenario, I'll use the remaining slurry to inoculated some grains or a brf cake.  Have a great day!

Keep it real sprocket ✌️

Has anyone done this? I want to do a bunch of transfers for inoculating grain jars with agar. My thought was taking my donor cup and adding an ml or two of sterile water to make an agar slurry. Then take an inoculation loop, dip it into the slurry and inoculate the other cups that way. Seems a lot quicker and easier than cutting samples out with a scalpel.  Any thoughts on this?

I think I'm going to try this except use a small syringe to suck up the slurry and then put several drops on agar.  Should be fast and easy. Thanks for the idea.

When I poured 40 agar cups, I thought to myself, this will last me a while....4 days later half of them are inoculated.  Looks like I'm going to have to pour some more soon, what a shame.

Can I get some feedback on if anything here is worth transfering?

Third pic, nice round colony at two o’clock looks really nice.

3 triangles out of that? And put mycelium down on 3 clean plates?

I would be tempted to grab a couple transfers from the plate 4 as well @ 5 & 10 oclock from the swab growth.  Plate 1 - the culture on the left has usable growth @ 5oclock as well.  Having more starting cultures will give you greater variety in the phenos that result - handy with nats since some cultures have overlay/growth issues.

Yeah, I'd personally go with the selections in pic two.

This is my first attempt... I cut the triangles with a sterilized blade and put them in a new plate mycelium facing the agar correct?

It doesn't matter which direction the myc faces, as long as it's quickly nestled in the center of the plate.

This is Pan. Jamaica. I see these little white specs almost like pins. They are under what looks like a fine layer of myc. Is this good to go? Sorry for the shitty picture.

Well, I used to me a member of this forum 20 years ago when I first cultivated GTs.  I had to stop because it was dumb for me to do it while active duty military.  I retired 5 months ago and am back at it.  Flow Hood and more equipment will arrive next week.  Agar has been inoculated.  Shopping around for a autoclave now.

What's the lighter for? I hope you are not using that to flame sterilize shit inside the SAB.

First and foremost thank you for your years of service!

To touch on the lighter. You want to flame sterilize you tools on the outside of the SAB. The flame can cause turbulent air movement and kick the nastiness back up in the air. Also if your using isopropyl alcohol in you box it’s a flame hazard. Welcome back!!!

I was flaming out of the SAB but just took the lighter in and out of the sab with my hand so i did not have to reach around the table every time.  Also did use iso in the SAB a little before opening the bags i had syringes in.  First time using a sab.  Back in the day i just made a clean room in the house.

I have a alcohol lamp and plenty of torches.  This is what I chose to use today.  I didnt feel like grabbing my torch in the shop while I was doing this because I was watching the WHL championship game. I am in the process of moving so my shits all over.  Thanks for the tip.
