I've got super thick mycelial growth on all my t4 transfer plates currently running, they're 13 days in. Is this normal or indicative of stress? Bad pics soz but my camera's not the best. They're vertically fat also. What are ye thoughts!

It's a premix I've had for ages but it's Malt Extract, Dextrose & Agar

Looks like healthy tomentose mycelium. I’ve seen that puffy halo on the outer circumference on certain varieties before, I don’t think it’s anything to worry about. What variety is it?

The amounts matter. I'd say this is pretty normal looking growth for a higher nute agar. If this plate barely has any nutes I'd be worried. Since it's premix which probably wont be low nute I think this looks pretty good.

Umm... just throw the powders and water in the bottle and PC? it will dissolve correctly without agitation?

i put cold water in the bottle. add dry ingredients. shake and pc 20min.

when i put the bottle in the fridge overnight with just nutes adding agar the morning after and then pc?  the agar comes out clear as glass if using lme.

Has anyone done this? I want to do a bunch of transfers for inoculating grain jars with agar. My thought was taking my donor cup and adding an ml or two of sterile water to make an agar slurry. Then take an inoculation loop, dip it into the slurry and inoculate the other cups that way. Seems a lot quicker and easier than cutting samples out with a scalpel.  Any thoughts on this?

Sounds even easier. My reasoning behind the slurry was to use a 5 or 10ul loop then transfer and streak the slurry a little to get more/faster colonization.

I think the cost of diminishing returns with the slurry is medium/medium-high. The difference may be observable, but I don't think it will be significant enough to warrant a second attempt. Let us know what you find out though if you do attempt for sure, be interested to know.

I just did 8 traditional transfers today, but still may give the slurry idea a go. If I do, I'll absolutely report my findings back here good or bad. Worst case scenario, I'll use the remaining slurry to inoculated some grains or a brf cake.  Have a great day!
