At what point do we think we have a 'good isolate' when having plates like that? Can you take transfers of that to many plates and call those plates a good isolate to use for innoculating?







Took a couple transfers of this once I saw the insane rhizo growth a few days ago. It just continues on like a freight train.

I have a handful of transfers to go, only at T2. I'm working to get at least 4 plates that are near iso and hope one of em is a gem๐Ÿ‘ Riptide is the only strain I'm working with to have that crazy lanky rhyzo on all the plates.:shrug:

So the general consensus is to take agar to T4 plates, gotcha. I'm as far as T2 on some of mine and they already look good but I'll continue on. They're from a swab but the plates don't like to show much sectioning.

Any reason to let your plates grow out for so long before transfering again? I can't see any sectioning with my noob eye, lol.

Those Riptide transfers ripped across the plate before I could get to them. I usually let them get to about this point before I make any decisions. Here's a pan cyan plate about to be worked with tonight.

Thereโ€™s really no right or wrong, as long as itโ€™s clean. If itโ€™s your first transfer and itโ€™s clean, you can send it to grain. Or you can do what I do with clean transfers. Let it grow out into plate pins and send each pin to grain. Fastest clones you can get. Only downside is a fast pin on a plate doesnโ€™t mean itโ€™s going to be amazing. Some of them suck, so itโ€™s not like grabbing a clone from a full grow.

Typo. Fastest clones. Normally, you do a full grow and take clones from that. A clone straight from a plate pin is just fast and you know youโ€™re getting genetics that want to grow and fruit faster than anything else on the plate.

Quick agar noob question: I sometimes see T1, T2, T3, etc for talking about agar. I assume we're talking about the transfer count since the first agar plate was inoculated, right?

I also have seen some F1/F2/F3/etc sometimes. What do those stand for?

'Filial', it denotes generation count.  So, starting from grandpa makes my dad F1, which makes me F2.
