No. Clones are taken from inner tissue.

I realize that but you're still working near surfaces that were exposed to open air. I just would not ever trust that to be 100% clean. While the risk is very low if your technique is good I would still do atleast one transfer to be safe.

Sorry to double post but here is a Tidalwave plin I'm quite proud of. I know it aint much compared to what I see posted here but it's my favorite plate yet getting back into things after taking a several year break from the hobby.

.......what?

Dude, with that logic we wouldn't ever get anything clean.

We expose sterile, inner tissue, in a clean / sterile environment, using sterile technique.

You're massively overthinking it.

A large majority here that clone never find T1 necessary for a clean culture.

This is why many go to straight to grain with tissue, and succeed.

I see your point and realize I've definitely been overthinking the situation. Like I said I've only done one clone at this point. Good to know.

Once you clone a few times it becomes apparent that inner tissue is safe.

After some practice it’ll click 👌

Lol you'd have to come at me a lot harder than that to offend me. You taught me something and I appreciate that.

Made up several plates. Attempted to inoculate 8 dishes with MSS. You can even see the spores floating on top several. It has been 2 weeks and not a single dish has done anything. No mycelium OR contam. Somehow I doubt my sterile technique is so stellar. After doing some MSS to grain, I Went G2A on these same plates and they took off very well. What gives? Any ideas?

Your agar could be too firm. Overly firm agar can sometimes stop spores from germinating.

Also, did you use a loop or just squirt some on there?

I have a similar mindset as Nuclear on this. Just yesterday I did inoculate a couple bags with T1 clones from the interior of fruits so I am waiting to see what happens but I got to say I only did so because the plates looked so happy.

I will update when I see the results but my gut is always telling me to make "just 1 more transfer".

I just took my first clone to agar as well.

IIUC, cloning will (hopefully) alleviate the multiple dehydratioin cycles I currently have to go through with each tub - as well as yield fruit that is more uniform.

My plan is to then take grains that are spawned to new jars while the rest gets fruited.

It will be interesting to see whether you get uniform fruits and whether they grow at a similar rate.

Re cloning, do you always take tissue from the stipe or is it ok to take it from the cap? I've only done a couple of clones so far but was wondering about this a couple days ago.

Anywhere you can get some clean internal tissue is fine.

If you filled a 5 gallon bucket with agar and split a whole mushroom in half then placed it on top of the bucket you'd see mycelium growing from both the stipe and cap ...lol

Makes perfect sense to me... Not sure how you'd do it without the bucket....

Yes.

Then ruin it with some holes just so you can decide you didn't really need the holes and then tape them over with monkey tape.
