I definitely get those spots, and I agree that they are caused by condensation. Other than being unsightly, they have not affected me in any way, so I don't worry about them.

I don't see anything wrong with those plates, but I'm not a PE grower. I hope one of them will chime in for you.

Anyone get tiny water spots on the surface of their agar sometimes? Like the size of a pinhead and completely invisible except when looked at the surface with a light. I can’t even get a good pic.

I’ve never found them to be a contaminant and distilled water has cut down on seeing them for me. My other theory is that it’s condensation dripping down from the lid and then drying on the surface.

Just wondering if others see this often?

I get this on the bottom 2 plates when pouring stacks of 10, but I feel like my condensation is more visually apparent. I use a wire rack to elevate everything in my SAB off the tabletop by an inch or so. Figured this is caused by the agar cooling quicker, similar to how I get condensation on the lid of my top 2 plates for similar reasons. Not sure if this is similar to your scenario or not though.

My Ape cultures have never yellowed like that. However it is not metabolites so likely the shadows reflecting in a mysterious way.

Seems I went backwards…. I am not sure if it’s the agar or the myc?  T3 looked uniform and strong.  My goal is to get a clone for each of these back to agar and then make LC.

What’s the best path for these - send any of these to grain, scrap any, transfer again?

Bottom left B+ ,
Bottom left & top right APEU I would send to grains

Thanks!

A lot of my plates have been getting this weird yellowing of mycelium lately for both APE and PEU and I dont know why. Its not weird shadows it looks more yellow in real life than on pics. Only a few of my plates are very clear white so im gonna use these to make transfers.

But still I think its weird that this is happening often. It cant be a small amount of bacteria causing some yellowing like that?

This one has the most yellowing and also had the slowest growth

ive used these for tissue culture in the cannabis field but never for mycology. Do you like them considering you have about half or less surface area?

I don't think that's why I like them, I like them cause I apparantly have no contam and if contam will happen it will probably be very few like the guy above me posted his experience was...

Maybe cause the surface is smaller and less vulnerable is why no/less contam, I dunno...

I do have a nice flow hood and my sterile tek is "casual" so science would say at least the cups are sterile...

Ive had pretty good success with those, just seems like extra work though, opening and closing lids. but then again, its better than wrapping plates

Don't plates also have tops that get opened and closed?

well when your working with two hands and one hand has a scalpel or whatever, unsnapping and snapping the lids back on was always the hardest part for me. plates have no snap lids lol
