It must be yellow mold season, been a lot of it around lately.

I always iso down my whole work station in front of the hood. I get paranoid there’ll be some bit of agar or bit of clone tissue or grain that I dropped and didn’t notice and it got moldy and as soon as I turn on the hood it’ll just be blasting spores over everything while I work

I honestly for about 2-3 years got away with never cleaning the SAB, forced myself once a year.

Once I finally got triched from being silly it's been hell. I have a tent with my SAB so inevitable. Have to shut tent off and wait a long ass time before I work in SAB. Dreaming of a tough shed/hood to work in.

I get contams from my hairy wrists, I should just start dawning tyvex sleeves. Never bad practice to leave plates from each batch out and see if contams were from the pour or leaving that agar open too long (?)

I spray everything, including the outside surface of my hood. Everything gets sprayed and wiped down, especially my metal rack.

Some PE clone and wild poplar oyster on deck.

I feel you, AKA. I clean my entire workstation before and after every session with iso.

~I do wipe down the whole station before every session with 95% ethanol and don't have any issues (unless I'm sloppy and user error occurs). I don't have a dedicated space and have to set up and break down the whole party each time.

Control plates are a wonderful practice if your having unidentified vectors cropping up with plates. Just open one up in front of the clean air and close it back up. If that doesn't go bad you know it is not your air or space at least, and the next step is to open one up and cool a flamed scalpel in the edge and close it back up.

I still enjoy the taste of humble pie from time to time.

Fuck yeah! Lazy gang! Good sterile tek > cleanliness.

My micro labs in college were huge halls with no masks on, bunson burner, and us bullshitting around plates. Man I miss that micro professor he gave me some super yeast when I approached him after class about if he wants some mead/moonshine. Was lesson specific about Saccharomyces cerevisiae
If only I was growing boomers back then...Hey Prof. why is this plate so organized compared to bacteria?

Damn I'm still getting satellites around the edges of my petris. Only ones I've used the wrapped ones are good..

:notsureif:

it's the gabbk spell, don't look at his posts or you'll get satellites

I finally fixed it!! I think it was the saran wrap, I suppose I got it dirty with the knife when I cut it (maybe I forgot to wash the knife ). And also got back spraying the sab and using a towel. Those things helped a lot

EDIT: I forgot you joined the flowhood club recently. forget about the spray and towel

I bought one those little usb vacuums for keyboards to suck out those lil millet grains that bounce between my pleats. I been doing my g2g over a cookie sheet. Has helped tremendously with those stray grains that like to go bouncing everywhere.

Mushboy, do you use the same roll of cling wrap for wrapping new plates vs used? I assume you probably do since you’re doing it all in the same spot but worth asking

How long do I keep the control plates for to see if they picked up anything?

APE germination after 3 days. This agar recipe does seem anecdotally to be more effective at getting spores to germinate quickly. I have some allegedly 6 month old mexicana spores arriving shortly that I'm curious to try out with this.

To inoculate from a print I always just scrape with my scalpel. I've noticed a lot of people saying they have to cool their scalpel in agar before scraping but I skip this and haven't had a problem with dead spores as of yet.

I like using a inoculation loop because I get clean 4 quadrant streak patterns. I also wanted more lab time using a loop for practice and principal. People told me they swore by it and so I said fuck it and tried. It’s been a game changer and I am happy the people here argued with me on it.
