Question about Sectors, Rhizo, and what is the best area to transfer?

I'm pretty new to cloning, and also isolation work on Agar, so any advice is much appreciated.

I have done some reading, and I understand that the ideal is to get an isolated mono culture, and grow that out, and if that grow is great..keep growing that culture?

Thing is, I'm not sure what is the best way to start isolating from sectors..

I've marked the clock digits on the plates, and feel that both plates look good at 12.

This is a PE first transfer from the initial clone plate, which had a little contamination.

These look like clean plates!

If you're looking for a mono you want the smallest possible transfer. Try using the tip of a needle to scrape a tiny bit of myc from the edge off the agar and transfer that. Even a microscopic transfer will grow out.

From what I’ve seen you’re better off just doing a couple transfers then going to grain.

The PE clone I did most recently grew similar to ms with the original tissue but on t3 it grew the tub in my signature where it was dense and they all looked the same.

Based on that it seems like by t3 or 4 you’ll have it narrowed down enough.

AKA isn't a clone super narrowed down? Does that mean material from a clone is still pretty MS?

I think it’s narrowed down but in the sense that there’s only 1000 options instead of 50,000 so it’s still pretty variable.


I pulled those numbers out of my ass so don’t take it literally.

Played around with all my random flours and baby jars. Not a huge fan of the visibility into the jars. Also the myc is so hard to tell if it's healthy.

Seems to be taking a long time to grow out compared to MEA or even BRF.

Made some much better looking plates with some pp5 jewelry rounds. Hoping I can recover some lions mane that keeps contaminating out.

I have a bunch of contaminated plates that I hope I was able to grab something of today not sure though. Might just have a ton garbage plates from these. we shall see :shrug:



I like the way that the contam'd GT and B+ plates grew.

Tried my 100mm Poly Pro dishes as no pours, unfortunately I didn't let them cool in the PC properly so they had way more condensation than I would have liked.

probably going to order one of those 500 packs from celltreat, those are my favorite by far.

Have a few more MS plates that I am hopeful for, but it seems like the myc looks way different than I am used to. Either that or its all contamination. Time will tell.

Good luck man.

I just had a hell of a time getting a clean plate for a clone that got mold sporulating on the tissue plate.

It took three transfers but I think I finally got it. I kept thinking it was good and then eventually I’d see a little spot of mold starting somewhere on the plate.

It’s tough once the spores are all over.

Mind telling me what kind of poly dishes you have and how deep they are? I tried using my 75x12 mm petris as a no pour but even with a thin layer they boiled over.

They seem good, did not boil over for me I tried to make sure that the temp stayed as close to 15psi as possible, and I wrapped the whole stack in foil.

Only complaint I had about my first attempt was the condensation. Other then that they will be great so long as they are reusable.

too bad they don't have a grip ring though, that makes it really convenient.

They are reusable for sure I've got some going on 5 of cycles now same brand. If you got to the manufactures website on the box they shipped in the are atleast 30% cheaper. I can pm you the site if you pitched the box

These first two are of some Pan Cambo Sandoze...I just thought they looked really cool (that symmetry on the second pic makes me happy)





This last one is some PE plates. This typical of PE? How it’s all light towards the center and gets more opaque around the edges. None of my other cube plates have exhibited this, although I’ve seen something similar on some Pan cyan plates.

The “floor spores” is because the PE syringe got clogged and as I was trying to plunge a drop of spore solution on my sterile cotton tipped applicator, the syringe suddenly unclogged and I shot the entire wad of spores out on the cookie sheet I lay on the bottom of my SAB to keep things slightly raised off the bottom. I decided to dab my swab into the puddle of spore solution and streak the plates anyways. Only 2 of the 5 gave me anything but it looks like cube myc to me.

I'm yet to see decent science to back this up. Loads of anecdotal reports and talk or isolates. Also talk of genetic isolation within a few transfers.

Many will insist black and blue that isolates take a bijillion transfers, often based on the visible presentation of myc on agar. . .

If anyone can link to science backing up either side, please post. . .

My personal favourite was a lab tech suggesting that by the time an MS swab has visible colonies, these are the strongest of the germinated strains as they started first, secured more nutrient, so grew quicker that the competition thus securing more nutrient. . . Not sure why there are all these folks suggesting there are thousands of strands still competing even 1 transfer in. . .

As for using visible as the only measuring scale, enoki in the wild grow very differently to most that are cultivated. I grew subs that I were told were cyans by appearance till I showed them a picture of the p.sub I cloned. . . Why does myc have to be different? Doesn't the orientation of the transfer impact how the myc grows? Some speculate that myc grows on the surface of agar only. . . My anecdotal observations contradict that. . .

Since there’s no scientific evidence supporting either one why would you doubt the anecdotal evidence?

I’ve never heard anybody say myc only grows on top of agar but that’s definitely not true.

To me it made sense that there would be multiple genetics based on how mushrooms work, then this grow was enough to convince me for sure. Same clone grown side by side, only one is an early plate and one was transferred a few times.

APE going crazy af, started from a MS LC.

Wouldn't it stand to reason that by taking a clone of a singular fruit you are already isolating?
I am by far no expert on the subject, just stating an observation that I've seen with my GT clones growing uniform rhizomorphic hyphae without having to narrow it down from MS.
