I'm only running like 3 (cube) varieties,  but 2-3 clones in each variety, and some mutant cultures/phenotypes I'm trying to stabilize... I stage them a couple days apart so I'm not overwhelmed all at once, and so I've ALWAYS got something to do... this way I'm not fucking with my boxes constantly lol

King o’s don’t require as much FAE and can indeed be grown in tubs as the stem is the prized part of the fruit as opposed to the cap.  Coral-like doesn’t sound good.  Reishi is another that can be tub grown as again, FAE isn’t a real concern.

@Caps - there are some decent king o grows on here done with coir.  I’d still recommend actual wood over coir...sawdust is free and HFP are cheap compared to coir.

Nah man, supplemented just means adding wheat bran to the sawdust 1:10 ratio. Honestly I think if you increase your spawn ratio you can skip supplementing. It’s a win win. You don’t have to buy bran just use more spawn and your colonization time will be shorter as well

Yeah, any custom hardwood furniture shop/manufacturer will practically pay you to haul off their sawdust and shavings.  HFP are Hardwood Fuel Pellets.  Supplementing them or any wood-based sub is as easy as adding bran at 20% dry weight and then sterilizing after its all been hydrated.

Yes coir is what I did. Even got 2 flushes. Literally put it in a cold dark closet and forgot about it for a while. If it's dark there's no real color to the caps which adds to the spectacle.

Thanks a lot to Caps, I have a 3 raging RW plates. I am getting a pack of 12 polypropylene petris and a roll of parafilm in the mail today. Going to try and pour some plates so my cultures seem more professional.


Do you need a laser thermometer for pouring jars? Does anyone just pour based on feeling(warmth) and looks of the agar?

Here is 1 of 3 RW no pour jars, going to take one jar and make a couple of agar to grain transfers and take another plate and do some agar to agar transfers.

Copied this from an early post.  I've tried a bunch of different methods, here is what works best for me:
Here is what I'm currently doing that kicks ass:
Boil water, let sit with candy thermometer until temp hits 120F.  Transfer to SAB with media bottle in bath.  The water bath serves two purposes: 1) Agar temp, 2) Heat sink in the SAB....My SAB is hot AF while I pour.  (120F is to account for temp drop while prepping tub, ends up closer to 117 through the pour).  If my media is hotter than the water bath I wait until the temps equalize to 120F.  I pour 20 plates at a time.  Stack em in 2 10high stacks and cover arm holes with Papertowel.  Let sit for 1-2 days.  This reduces the mass temp swing that causes condensation.  Plus the condensation can escape the plates a bit.  After day two I prep for all transfers, remove heat sink (because it is room temp now).  I still end up with 2 plates that have condensation.  I set them aside and use last.  By the time I am all done my plates are clear as a bell. 
The hot cup on your plate won't hurt anything but the condensation will come back....you are essentially returning excess H2O to vapor...when it cools it will probably condense a little again.  No biggie as long as it doesn't run on the agar surface. 
Room temp helps too...I'm working in a mini-lab built from a 20ft shipping container.  I turn on a oil circulating heater for the pour and while it sits to keep a solid high 70's room temp.
Seems to work for me:)

cool, ive seen really nasty shit and trichoderma in my first agar tubs before i picked up the pressure cooker, most where so bad i threw them straight out without opening haha. Got paranoid after spotting those orange blobs on a mature plate, checked all my tubs and cant see anything out of place :laugh: the Bacteria not fucking everything up with spores is very kind of them

i use Pasty-like tubs, pouring them 1st then sterilizing works fine but too much condensation for me.

I sterilize the tubs empty at the same time as the agar, leave the tubs in SAB while agar sits in water bath down to 45C/47C then pour quickly, still condensation but much less :smile:

I broke the no spill plastic ring from the media bottle :'( few drips now ha

Could I do agar to grain if my agar is only 60% colonized? I could still use wedges from itnim sure right

SR, I learned right of the bat: pressure cooker and SAB...shit, I built a glove box that I thought was tits.  Then, got slapped around a little on here and modified to SAB.  I'll never go back!  I've been relatively lucky with the agar experience as I've had lab experience, but I still started out with condensation on my first 20 pours trying to adjust to SAB vs. lab flow hood.  I use my PC extensively as an autoclave too.  I have my scalpel handles, and tweezers cooling in the unit now for agar work later today.  I wrap in foil, use some autoclave tape, PC for 45min at 15psi.  Voila, sterile tools of the trade.  I don't have to bring home boxes of throw away scalpels anymore and I'm sure the environment loves me.  Keep posting pics of progress.  Would love to see another newbie to agar progress for note comparison of what works and what doesn't!

I  just wipe handles down with a wet paper towel, and flame sterilize the blade... or wire loop/tweezer ends

I want to clear up a couple things before beginning next grow.

I am going to be using a MS Syringe from a vendor.  I am using the pastywhyte tek to do it.  How common is it to get an agar wedge w/ no contaminants?  If I have 5 dishes and get a spore drop on them is there still a high probability that I will need to do at least another transfer before putting in LC?

I want to get my expectations right on this and through reading I might also knock up 1/2 pints of rye and transfer a piece to each dish as it seems it may be better than the syringe route.

Make a few transfers from germ plates to ensure you have a clean culture... it's why we use agar at all

That’s probably all that’s needed but I have to stay in a workflow routine. Stick with what you know, and all that. The routine of opening sterile autoclave package and setting up sterile-field  equipment station gets me mentally prepped for the work. By the time I’m on the agar I have surgeon steady hands and I’m ready to rock. I do the same routine in my other life and it calms and focuses me.
And now the waiting begins:

Hi guys, noobie here. These are the 4/6 plates of PE from a spore syringe that had some growth. 2 are contaminated. I was hoping to get advice on which ones/sections to move to another plate. Thanks!
