I'm curious if anyone bothers to pH the Agar? Does it make a difference in growth?

JJ

I do when I know i will be using it for spores. Spores can be fussy about pH. Once they germinate the mycelium can take wider pH range.

Recent clones making good progress!  (Can already hear some comments.  “Why did you take so much tissue?”  “I usually take 1/4” at the most”)

My answer - That’s just how it worked out.

1.) These are mini pin clones I took from this Roatan plate.  (Didn’t save the actual pic.  This one is from about a week before I transferred).

Best looking plate:

2.) Roatan clone #2 - Cloned this fat boy.

CP1 -

CP2 -

3.) AA+ clones taken from this specimen.

CP1 -

CP2 - (**Changed pics to actual AA+ clone)

Thanks!  I’m pretty stoked about them.  No bacteria or mold.  I think just about every clone I’ve done in the past has had at least one plate with bacteria right off the bat.  Guess I’m starting to get the hang of things:-p

Note:  I put that disclaimer in just for you Caps :liottalol:

So I accidentally made agar watery with some psilocyb Cyanescens. Went from print to agar. Third transfer. Should I toss it. I haven’t seen any contamination’s yet. Been about 5 days.

Also here is 3rd transfer from a wild oyster clone I found. The agar was also watery. Are these all just mycelium points or is it just fucked.    Sorry for the crap photo. Pasty plates are hard to photo.

Well idk why you have growth on the wall of your pp5, bu if it's clean then transfer it again and do it right.

On the 'best looking plate'... did you drop two tissue samples on that agar?

If anybody is willing to burn a little time with suggestions today, fire away.  I tested an LC for contams to a couple PF TEK jars I had left over from another project on 1/5/19, birthed the cake in the pic 1/25, and now woke up to decisions to make.  My previous tissue to agar samples did not go well and I think part of it was poor donors and shitty agar pours.  I now have plates that are money, clear as a bell and without the gallons of condensation of my previous disasters.  So, obviously the cluster is the best choice, ya?  Do these fruits express traits worth cloning...to me they look dope, just not super huge.  Which specimen would you take tissue from? And who prefers what, base or cap meat?
B+ from LC to BRF

I don't want to waste plates, and have plenty of other projects, but if these look good enough let me know.  I'll pull 2nd and third flush prints for my library and trades.

So pretty much everything i got grows like this for a few transfers. Is this some type of mold?

I only ask cause recently i changed some things up with my agar and the way i work and now it looks so much better imo. But if they are both healthy then who cares.

I'd clone two of them. On the first pic, the one at 10 o'clock and at 2 o'clock look good to me.

The bottom third of the stipe is my usual place to take the biopsy for cloning. And remember that less is more in this case, less then a grain of rice is best.

No.  I took several of the mini pins, avoiding tissue, to clone.

This clone plate is looking so good I might let it incubate for a couple more days and transfer to grain.  Bad idea?

Bad idea. While the mycelium looks good, you do not know if there is a spore or few of contaminant on top of the original biopsy piece. Move it to fresh agar now, it looks ready for that.

Right on.  Yeah I was going to transfer today but thought it may be worth transferring to grain.  Was just a thought.

Really like how this Caerulescens plate is coming along.  Looking more aggressive than previous cultures.  Almost looks like Cyanescens myc.

HH, I'd clone at least the largest of the cluster, just to have a clone culture of the variety on hand... remember that smaller tissue samples will recover more quickly and carry less risk... BB, I've dropped a few tissue plates with success, just depends on how confident you are with your sterile technique when it was taken... good idea to transfer a very small wedge to a new plate for expansion: just in case... alternatively, you could drop and omit the center wedge with the tissue sample on it entirely, just flip it out onto the floor of the SAB with the scalpel prior to A2G

Yeah, I wasn’t going to transfer the clone tissue.  Just the myc.
