LOL funny story, my main work table is made from a solid core door.

Anyways... I feel like white would make it harder to see the edge of new myc growth? Am I crazy on that?

Mycelium and other growths are best viewed in a reflected light. Coloring of the media is immaterial when you are using it as imperfect mirror.

This is a great idea... however I'm only using a SAB, do you think it's still worth it to put that in the SAB? There's so little room as it is.

Good call...I’m relatively new to transferring but I’m on PDA which is pretty tan and I have killer led lights in my workspace because it is my HIFI equipment build bench. I find myself hunched over the SAB peering through the armhole while I work. I use glass dishes flipped over as the work table in the SAB to elevate off the “floor” of the SAB with paper towel on top. I’m able to see the edges very well from the tan contrast of PDA  on white paper towel.
AND AS CRAZY AS I AM...I definitely can’t speak to your level of crazy:) But I think your reasoning is sound I just haven’t experienced a problem with seeing the edges myself.

I do not use it in the SAB, I look at my dishes before I do the work, and if there is a small feature I will need to see when working then I use a marker to highlight it on the bottom of the dish. That is rarely needed however, because once you look at it closely it will be much easier to recognize whatever you are looking for without the magnification. And again, have a source of light near and slightly above your SAB so you can look at the dish in reflected light. That really highlights everything.

GENIUS! Actually, for me at my low level experience, this will be super helpful. Esp. since I feel like I'm struggling to see ANYTHING in my SAB most of the time. I do have a light I put on the top of my box when working with agar.

I'm waiting on a HEPA filter to build a flow hood, so am in a SAB for now... once your plates are wrapped, you can play hot potato in a dust storm if you want lol... probably be cleaner to use the hood and transport... though if you're having success in a SAB,  I'd not worry to make it a regular thing

I also use a marker on the bottom, but for contaminants... I have a dome light over the SAB and hold the plate up to the light while peering through the armhole to view for choosing transfer sites... I've seen some guys lay a light behind the SAB facing them... mine's a bit bright for that though

I like the color to be able to differentiate different agar sessions at a glance. Like peek at the shelf and remember that blue has all my t2 transfers for a certain species and orange is the antibiotic agar for t1 of that other species and so on. 

Also yeah I’ll be cooking my agar for at least 5 min for no-pours. I feel like it didn’t solidify bc of rushing. I had no hard sediment or alcohol in my dye- I think it was just visually to the point of looking combined but the agar hadn’t activated. It did a little bit of weird coagulation after sitting for a while, and it may have set up after PCing, but i feel that there may have been different concentrations of ingredients in each plate had I gone ahead with it.

Well this is my first time attempting with agar. I just poured my plates. They're sitting in my SAB at the moment.  But I need to put the SAB Away (takes up too much room). Anyway, I have an extra shoebox. I was wondering if I could store them in there until I start making my clones?

I have some parafilm.. I was going to wrap them.. but is that really needed? Can I just wait until I drop some clones in there?would hate to wrap them all just to unwrap then rewrap

Wrapped dishes are safe from contaminants: it's why we wrap them... open shelf storage is fine

As soon as they are set you can put them in ziploc bags. A regular sandwich bag holds four plates. You can keep them at rt, and if you got any contams during pour they will show up before you use them. Or you could stash them in the fridge for longer term storage - that will reduce evaporation. Either way works.

unwrapped and undisturbed, contamination won't be a problem, but they will dry out after a while. as alluded to, if you use them soonish, no need to wrap them. if you're gonna need a few weeks, why did you pour them?

I'd transfer from around 11-12 o'clock; the little white bumps are yeast colonies... wouldn't hurt to let it grow out another day or two and see if the growth at high noon stays clean, and can get a little further away from the undesirable fungal infection... take a very small wedge (rice size or smaller)from the very leading edge: it only takes a microscopic bit of culture to inoculate a clean reciever... hold them up to the light daily to look for dark/thick spots in the cultures... otherwise healthy mycelium can overtake a small contamination and hide it from plain site rather quickly : especially tomentose growth

So i put 9g pda to 250ml water, my plates were washed with antibacterial soap then pc'd for 120min, agar pcd for 12min +/-3. Before innouclation (p.mexijal) i cleaned the scapel with 70% ipa then air dried, no flame sterilization, stabbed about 1mm into agar... And 24 days later i got this

Bunch of nothing not even streak of some trich or cotton... my question is has anyone ever had spores take more than 24 days to germ? Please any thoughts before i  :alphabrain: and slam this shit in the garbage?

Iso doesn't kill spores... just so you know

Spores can take up to 6 weeks to germinate... 3-5 days is typical IME

Even trich will take days to show: hell, a transfer from a living culture takes 24 hours to recover... patience dude

I'd probably streak the surface a short distance rather than stabbing
