Preferred Agar Recipes for Huatla

Trying to find preferred agar recipes for huatla. I tried PDA but for some reason the ms syringe didnt take when i did a swipe (the golden teacher is loving PDA). Will try some LME when i can get to the brew store. I also tried to put a swipe from a wild print of Pan Cinctulus on the PDA with no results...

Right now Im using pastyplates with 500ml water, 10g agar powder + 8g potato flakes + 6g corn syrup

Huatla is a name printed on a label it has absolutely 0, nothing, zilch, to do with the agar recipe you choose.

Cubes work great on PDA.

Trying to get spores to germinate on agar tends to need a slightly less hard mix, at least in my exp, it's a little more moist etc..as you're currently using 10g, that's gonna be quite hard, so try half of the usual amount of agar i.e 5g and test. You could cut the potato back to 5g too I'd imagine as that's the amount I use with good results

I use the same mix for everything. 2% agar, 2% malt extract.

May be different for PDA mixes? I've had zero % spores germinate on 9g agar, 5g Pot, 1g yeast and 10ml Honey, from spore prints or syringes, even though normal transfers love it and cover a plate in a few days flat..Couple of old threads that had good ideas in came to the conclusion that it was the agar stiffness

You're adding yeast? It's supposed to be yeast nutrient, not actual yeast.

I've used PDA before and never saw a problem.

Yup actual yeast, just ground up into a fine powder rather than the tiny round balls....Seems to work as growth is rapid, I've been thinking about giving Vegemite a try to see what happens

I hate seeing recipes like this in percentages. is it 2% by weight, volume, also with water and powder ingredients it gets confusing. This is what i use which is probably similar.

1 teaspoon agar
2 teaspoons potato flakes
1 drop karo
1/2 cup water
1 drop food coloring

Mix together in a sauce pan on medium. Pour into jars or glad mini rounds and pc.

Vegemite is all salt so I would suggest avoiding it.
Also yeast should be autolyzed yeast extract for agar

Spoonful recipes like that are atrocious. Percentages are ideal

Guess I just suck at math, but percentages confuse me in agar recipes. Maybe it's better for larger amounts. What i posted makes about 8 glad mini rounds.
