Agar recipe work well for germinating these five spores?

My next door neighbor has the worlds ugliest hooker that comes to visit him. She (he) tells me about her (his) experiments;

She was making some Agar, MEA actually, from the Agar FAQ.

She put the 500 ML of water into a jar, and nuked in in the microwave for 6 minutes, got it all nice and boiling.

Then she added the 10 grams of Malt Extract, 9 grams of Agar Agar, and 10ml of honey, just for fun.

This got mixed real good in the jar, then the jar spent another 3 minutes in the microwave on high, then another 10 minutes in the microwave at half power.

Into a glove box she put her petri dishes, then the still burning hot Agar jar.
She had hosed everything down with bleach solution and the alcohol-mist, washed her hands and her cooter really well, and then poured the Agar into the petri dishes.

She had never done this before...at least not taking it this seriously. She had experimented with playing with Agar and a little honey and barley malt before, but not with petri dishes, she used some other kind of container. The chunks of mycelium on those agar "plates" is actually growing, but it seems really wimpy. Maybe she isnt singing to it enough?

Now her questions: Is that microwaving technique going to be sterile enough?
Will that Agar recipe (10 grams Barley malt, 9 Grams Agar, 10ml Honey, 500 ml water) be good for germinating some spores? She has exactly 5 spores, she counted. Not many spores and they are the last 5 spores in the world left from an extremely rare and now extinct shroom, the spores where scraped carefully from the shroud of turin, and are from the magic mushrooms that Jesus and his disciples ate at the last supper. (They must have been good, considering all of the wierd things that happened during that party, and right afterwards....hell, those guys even started kissing each other!).
So it is fairly imperitive that these spores germinate and form mycelium.
So will this Agar recipe work well for germinating these five spores?

How should she go about 'planting' the spores onto the Agar? Just place them there one at a time? Make a very small spore syringe?

And what does Agar taste like?

You're going to have to make sense if you want helpful answers. 5 spores? You can't have 5 spores. What do you mean?

I think the question was about the microwave sterilization tek.
Will that work for making Agar?

The other part of the question was about the agar recipe (listed above) and if there is any agar recipe that is better then another for germinating spores and growing out mycelium for transfer to H2O2 agar?

She has a very small amount of spores from a rare and hard to get shroom, and these spores are probably contaminated, but due to their rarity and religious importance, she feels it is worthwhile to try to resurect this shroom.

My neighbors ugly hooker saw a box of nutrasweet.

On the ingredients label it says "Ingredients: Dextrose with maltodextrin, aspertame.

Would this work to satisfy the dextrose requirements of some of the agar recipes?

Just for fun....Either you follow a recipe and you will have success, or you don?t...

There is no word about honey in the MEA recipe....

Well, heres what she was thinking, after reading these agar recipes in the FAQ:

Malt Extract Agar (MEA)
10 grams light malt extract
9 grams agar agar
500 ml potable or distilled water

Potato Dextrose (Yeast-extract) Agar (PD(Y)A)
broth from boiling 150 grams sliced potatoes
in 500ml water for 30 minutes(add water to 500ml)
(You can use 5 g of instant potato flakes instead)
9 g agar agar
7 grams dextrose( = glucose, or 10 ml honey or corn syrup )
1 gram brewers yeast or yeast-extract (optional)



The Potato Extract Agar listed honey as an ingredient, presumably as a source for dextrose.
So she decided that a glop of honey in the MEA recipe might be a benificial thing....She's always got to be screwing around wth stuff and "making it better". So your point is valid "Dont screw around with things that are known to work".
But do you think that the addition of honey to the MEA agar recipe was a BAD thing?

And what about this nutrasweet stuff? Is that a viable source of dextrose?

What are the contributions that each ingredient make to the over-all recipe? What is it that yeast does for some recipies?
The agars purpose is obvious: It provides the gelling properties.

What are the things that mycelium and/or spores need to grow or germinate?

I appreciate your answers and input!

KingCobweb, it could be a good source of dextrose and maltodextrin for agar formulations...

Yes, because you are adding too much sugar.
The malt extract contains mainly malt sugar.
No need to add even more sugar in any form.
