I only have BRF cakes on hand right now, no grain. I'm going to make the LI in a few hours and suck it up into 2 60cc syringes and 10cc into a recycled spore syringe for my 5 1/2 pint jars. I figure 2cc per jar of this stuff should have some pretty fast colonization times, assuming I did everything right.

I'll update the thread later for sure.

I transferred the quarter-sized mycelium wedge into the jar and used my hand drill to chop it up, but it's barely working. I don't know if it's my drill being too weak or what, but it's not chopping it up much. There's little chunks everywhere, but there's still a dime sized chunk of the initial agar wedge in there. How blended should it be?

Edit: Here's some pictures. Should I keep blending until the bigger chumps have blended or do I risk harming the already blended mycelium?

It may have to do with the larger jar. That's why I like 1/2 pints. They hold the agar tight against the blades so that it gets chopped up rather than just moved around. Also I think you need to run your drill in revers to get the blades to spin in the right direction.

Yeah, I think it's the jar size as well since it allows the stuff to run around and duck away from the blades. I'll run it in reverse and swirl it around a bit more. Should the agar itself be completely blended, e.g., no pink anywhere?

I don't think I can get it more blended than I already have without damaging the already blended mycelium. My blade is hitting the side of the jar once every while too. Is it worth trying to suck up now, even though I don't have a completely uniform blend?

Ok, thanks. Yours looked a lot more blended than mine. I'll suck it up into syringes now.

Any objections to this:

I plan on sucking up the LI with 2 60cc syringes (the sterile ones in the package) and about 5cc in an old spore syringe. I plan on cleaning the old spore syringe by sucking up and spitting out boiling water a few times. Then I'll wipe it off in my SAB with iso and flame sterilize the needle before sucking uip LI.

I'll do the same thing with my sterile 60cc ones, minus the boiling water part since they're already sterile.

Ok, sucked up 2 big syringes worth and a small 12cc one. I inoculated 5 BRF jars with nearly all of the 12cc syringe, so they should colonize pretty quickly, unless there's such a thing as using too much inoculant (perhaps making it too wet?).

Edit: I just found this quote from eatyoualive on one of the earlier pages:

I had a tiny bit left so I inoculated 2 Pasty plates as well. With the remaining amount, maybe 1/2cc, I dumped it into an existing LC that never colonized. It was with the VERY end of an older spore syringe so I don't think the spores were viable. I don't anticipate this LC to produce anything I can use, but fuck it, I want to see some shit grow in that damn jar.

I'm hoping my methods were sterile and I can see some quick results with this stuff!

Quick question: I don't have any Ziplocs big enough for these monster 60cc syringes. I simply wrapped them in tin foil and put them in a paper bag in the bottom drawer of my fridge. Poifect?

thought this could add to this thread as your post a while back inspired the thought :cool:
so here ready for the first wide mouth LI Inoculation and just wanted to show it off. This would make easier transfers and possibly even a tiger drop with the ziploc rounds. It was an accident originally when i overheated my lid while torching it to attach stiff felt via dilated's no silicon lid tek and ended up with a wide open hole that would just fit the blade through. SO i decided it was destiny that i figure out how to use it with the oster blenders. THEN I meticulously started loosening screws on the blender base and soon got fed up with that and took a pair of some giant heavy duty pliers and went at it. I just broke the plastic rims off from above the blending rod on the base and it works great. THEEEEEN I realized i could have just used a drill to blend the jar. OH WELLLL. Then i siliconed the blade attachment to the lid on both sides
SO it's clones to pasty plates tiger dropped to MUDUFUKA's LI to cakes (or bottles and now invitro slurried clones from cakes straight to cakes and bottles) to eatyualive's slurry to mudufuka's bottle tek. We'll see how it goes....

The plastic lids do not form a water tight seal in case you don't know.

I also find quarts a bit too big. Narow 1/2 pints holde the agar closer to the blades. So it gets chopped up rather than just moved around.

the plastic not forming a water tight seal is debatable:
NO LEAKS
But i do agree as far as LI the smaller jars are better but with my wider ziploc rounds the transfer of large wedges  will be much easier and makes the tiger drop available. I have half pint wide mouth jars i will be trying with today...

it is debatable but the leak may be very small, just a drop or two escaping.  I'm not sure it's worth worrying about but with metal I don't have to worry.

use plumbers tape around the teeth of the jar before u screw the lid on and PC for LC/LI and plastic lid

I think the blenders ruber gasket makes the seal more water tight than just a plastic lid on its own

oh yea, forgot about the gasket, ur right.
i gotta get back on the LI train.  haven't used one in a while.  i wasn't able to use all the active grain spawn in time, and it over consolidated, so i chilled out on it for a bit.

ok i think i see a drop leaking through. Like you said it is a very slight leak. I peeled her off and made up a metal one thanks INVITRO!!!
I may try plumbers tape on the one that is still plastic so i can use it today as i was planning. Thanks for stopping me before i got in too deep.
