10cc per pf jar??  u crazy mofo!!

i will say, ur brf>grain thread is the fastest leap off i have ever seen, even faster than commercial oysters.  I'm sure it would be faster than petri LI as well, but i still don't see a brf jar colonizing in 3 days, no matter what the innoculant.  ill have to give it a try tho!!  i never write something off until i try it.
but i do like the brf slurry>bulk idea.  when i started bulk, my brf>coir was faster than grain>coir at the same ratio, and all i did was hand crumble, so breaking it up even more would increase speed id guess.
for grains tho, i still think petri LI's are easier and more efficient in the long run.

I thought about using a syringe but so far I have just poured the LI into each jar. All I have are 10cc syringes. I would like to get something much bigger before I star using syringes for LI inoculations. Pouring seems to work pretty well for me though.

i was just reading tv casualty's slurry in a hurry tek from a while back and looking at the picture progression of the 3 day colonization. what ill do is update my slurry thread to show the jars day by day with some pictures. its probably been around 2009 since i tried this method. but it works well. and many people have used it with good success. ive also seen people use it with straw logs with oysters!

yeah why add the extra step if it works? but you could make syringes to store in the fridge for backup when you want that clone later on. so you make 3 syringes for storage and then inoculate the rest into jars!

then you don't have to start from scratch to isolate it again.

yea pouring is faster.  this tek will shine with grain bags too, because pouring a little more by accident won't throw off moisture as much as it will with a qt jar.  i used to put a thin layer of dry verm at the bottom of my grain bags anyway to absorb excess moisture, so thats always an easy fix to a potential problem if someone was concerned about throwing off moisture content

that would be dope!!  i don't think petri LI would work well at all with straw logs for oysters, but slurry in a hurry would add the myc and the extra nutrients.

yes. even with grain jars there is room for error. because you can shake up the jars to redistribute the moisture if you see it pooling in the bottom. although with the slurry you get worried at first because the slurry pools. but it colonizes so fast that there isn't an issue.
yes i meant that the slurry was used with the straw logs.

I plan on using this mainly to inoculate sterilised substrates for  bottle grows skipping the spawn run all together so throwing the moisture content off a bit doesn't concern me as much.

here is a good pictorial for the 3 day growth used with the method i mentioned above. one is tv's test run the other is my test run. also there is a spawn bag with the same method.

also here is a spawn bag used with the same method.

Here is a test run I did on the same method.

ive also grown out those quart jars to bulk subs with no contamination.

here is a sample of the liquid being poured into a bulk substrate. fruiting conditions weren't optimal at the time but it worked. i think i also dropped the bin and broke the substrate in half by accident.

No it's in jars. I plan on putting them in my GH once they are fully colonised. I will likely switch to bags once I get a bigger PC. I am saving up for a 150qt.
For now I think the bottles should be fine. side pins are a bit of a concern but we shall see how it goes.

I also plan on making some jars like this using heavily supplemented sawdust for my enoki and oysters.

nice. i followed tv's tek but i actually did the method less clean than he did. i simply did what you described and just lifted the lid and poured the liquid inoculum into the jars/bags ect. he actually uses a built in nozzle that helps measure evenly the distribution into each quart. i didn't feel like building it at the time and my ghetto method worked fine lol. did you see the picture of the drip nozzle attachment he built?

this could be useful.

this is also another idea that could be useful. you would have to figure out how to attach it.

not to raid the thread, but just to illustrate that you can use other media than agar slices. all of which use distilled water only.

you can also use this with pf cakes or sliced stem tissue. you use a larger small mouth pint jar and it fits on the end. then you can blend it up and pour it into grain jars. using liquid inoculant.

ok  so do you just pour sterilized distilled water into the pint jar with the cake in it. or just blend the cake as is? Sorry i'm not fully understanding the process but very interested seems way to good to be true...
I have some cakes nearly colonized at the moment and would love to expand them to that extent. Oh shit just realized i can't since the blender attachment is not on that jar,DUH
so glad you said something about oster's plastic melting in pc. Do you think if I got the white model it would have been pcable?
what do you think of inoculating brf cakes with agar LI to make it colonise very fast ( since i'm scared of the tissue sample method) then use that as said above to inoculate qts of grain?
biggest question is how do you get the cake into the liquid?  :wow:

Wouldn't using LC kind of ruin the whole point of this TEK?  Seeing as how this tek is useful because it WOULDN'T support contams, but if you used LC then you would just be negating the benefits of this tek IMO

Oh, and good job OP.  My agar equipment is arriving on monday,  and this may be one of the first things i try when i have a good isolate/wedge.  Thanks for posting

umm no if adding the liquid to graain jars then any contams will show up there. If you have sterile technique down then there should not be any more risk it;s just a waste of time it seems. I don't know if this is what you meant but if you were to have poor sterile technique and got minimal contam while inoculating, then the LI may have better chance (not much) because the contam may only be passed to  some of the jars whereas a lc could get contammed while innoculating with wedge and (while colonizing) spread through the entire LC making so no jars would be clean. IMO:shrug:

oh yeah just gotta say i didn't think this thread could have gotten better then i read all this. You guys have some great ideas thanks again

thanks eatualive for the link missed that...  and now back on topic: First off i used the Oster blade assembly with the jar ring to hold it as eatualive's link shows. I just left it slightly loose as muda did with his for ge> SO glad i didn't pc those blender bases. Now  i have my first two qt jars half full of tap water for LI  :headbanger:  sterilizing at the moment and was wondering if you thought i should raise the pc time a few minutes to account for the extra volume? I added tin foil over the lid as a habbit and precaution but could that cause any problems?
