You are talking about selectively breeding with spores. The only way to absolutely guarantee consistency is to do a tissue culture and maintain the strain that way. Everytime that you go to spores it is a crap shoot to some extent on what you are going to get. If you want to maintain a potent strain, learn how to do cultures on agar

I know that.
I mean take the mycellium and let it colonize a larger substrate, and if by chance you get a stronger cake in the other terrarium, replace the old colony with the new one.

That would work but I would suggest spreading it out into several containers as well as several slants. That way when the cakes play out you will be able to go back to the original strain

What you need to do is seperate about 30 unique mycelial germinations before they mate and then do a lot of mating attempts (not all will be compatbile) and then take the petris that show fused mycelium and run it through a lot of bulk material to do strain analysis and well you may end up beating the odds and getting a good chromosonal replication mutation and hopefully breed it out long enough to favor that chromosome sequence.

Its actually quite a lot easier than you think :-) Gotta do the homeopathy tek of dilution ten fold and you can pretty much get down to a spore or three per ml of water.
