My office is video booth #2 at the “The Pleasure Chest”. You can check in with my secretary. He will be the one in a biker jacket and ass-less chaps.

That might be bacterial, although it is hard to tell from the lighting. Can you take better pictures? If it is bacterial it may or may not recover well after shaking. If it is healthy a shake won't hurt, but I usually wait until 25-33% colonization.

Shake it when you feel like its colonized around 20% or so. I think thats consistent with what I heard hamloaf say the other day which is probably about when i shake.

You're good bud. You're doing fine.

Is a proper pour bottle with the drip ring mandatory for pouring agar? Can I get away with doing it with just a grain jar or a regular glass bottle or will I be making a huge mess and contaminating my plates?

What’s the consensus? I could order the real bottle but won’t make it til Monday or so and I’d love to make some agar o. The weekend. But at the same time I want to do it right haha

Just use something with a small mouth and ideally a neck leading up to it so you're not sloshing everything out too quickly. I feel like I've seen people use kombucha bottles before.

Wheeeel,

Ive seen people use pp5 containers of all sorts, but ive also seen people use whiskey bottles etc and have them explode in their hands.

Heres the thing to remember:

Normal glass is good for big temp range, but NOT rapid temp swing.  You dig?

Borosilicate is made to be safe for rapid temp swings.

Id rather see you find a pp5 solution than burn yourself horribly.

Can it be done? Yes.  Have people done it for years and gotten away with it? Yeah probably.

Will i recommend it? Heavens no.  Same reason you wont see me recommend people microwaving agar etc... im kinda programmed to give the safest answers possible.

Solid advice thanks guys.
I hadn’t considered PP5 that’s a good idea, I’ll go to the shops today and see what I can find.

Yeah I’ve burned myself badly before in the kitchen and I’m not a huge fan. This was pan frying an orange roughy back in the day. Still remember the relief of having my arm under cold water when it happened, and the instant burning sensation when not under cold running water.

Yeesh, didn't know people had them break in the hand during pours, just seen multiple people do it so figured I'd relay that much

How does everyone store their long term culture?

Castellani tubes?
Slants?
Agar plates in the fridge?
LC in the fridge?

I've seen so many different methods. My slants SUCKED and didn't stick to the tube. So I ended up running the Castellani method with distilled water and stored in the fridge. But I want to know everyone's take on long term culture storage. Outside of prints**

Slants are ok but you shouldn't rely on them for more than a year. The fridge is useless. It's not true cryogenic storage. Castellani has been proven to be the most reliable method for us hobbyist. Try not to rely on things staying good forever though. You WILL lose cultures and some of them will be your favorite and it will break your heart.

I just switched to cryo vials. I’ve had cultures die ( plates) in the refrigerator. Takes way less space too.

thanks Pheno!!! I just did it all with my best cultures going to Castellani Tubes. I figured those would be the best "Long Term" storage. I figured nothing lasts for every, but those would be my best bet since they have a lot of data behind them.
