Year old dried out plate and pin. Any bets if the fluff on the cap is mold or myc? I'd myself give better odds to pure mold. Though that didn't stop me from picking it to a fresh plate, also took a piece of dropped spores to a new plate.

Aww thanks bud. But to be fair I bought those pre poured plates. So... kinda cheating ahaha. I did the transfers though.

I will be making my own agar soon, waiting for the plates to be delivered and then to have the time and energy to go for it.

So I'll be streaking some plates tomorrow using the sector method.

Has anyone switched to this method and noticed plates were easier to clean up (faster), as opposed to just a standard plate streak?

And I'm assuming less is more with spores, right?

We could totally put a big clump to agar, but at the risk of a higher chance of contamination?

If anyone happens to have any tips on the best use of spore swabs, that would be great too.

I like the idea of burying, but getting my swab handle short enough, and cleanly, is the problem.

Sectoring is more for isolating monokaryons through serial dilution, doesnt really have anything to do with contams. The less germinated spores contributing to a culture, the less genetic diversity it will have. I know some folks like to use tweezers to rip bits of the swab off for plunging into agar or BRF pucks so you dont use the whole thing in one go

Thanks for the reply dude.

Agar Plates and Removing Hidden Contaminants

Prevention

The best way to have clean plates is to never put contaminants in there to begin with. The way we germinate and select viable colonies can prevent the enmeshed bacterial contamination we fight with. Reckless spore streaking and wanton scalpel work brings along the very thing we don't want.

What we want to do is thin down the concentration of cells allowing for separation of growth. Separation facilitates isolation. Without the proper separation between cells, one risks growing the mycelium on top of yeasts, bacteria, and molds from the very start, making it incredibly difficult to isolate a pure culture free from contamination. That goal being difficult when starting with spores because of their nature, we can still minimize the amount of crap we take out of our germination plates on our first transfers by using proper streaking techniques.

I have seen the pulling threads technique, but I could see it getting stuck to the tweezers.

I guess I'll just try a bunch of different ways.
