Somehow my instant pot ran dry last night and shut itself off at some point. I have no clue how far it made it through the cycle. I'm thinking it was like two to two and a half hours.

They were no prep Millet jars. They've still come out hydrated. So do I just need to toss these things and start over or do I stick them back in the PC and do another run?

Lmao no worries.

TLDR: I turned 10ml of vender LC into 600ml LC and store it in a fridge to use months later. is this a bad move?

A lot of the ogs here who use LC treat it as a single use item, so having slightly less mycelium in the liquid doesn't really matter.

So being a noob, I took my vender purchased 10ml liquid culture syringe and put a few drops across 3 plates, ~5ml into two LME LC jars, and the last 5ml into grain.

Now I have north of 50 10ml syringes of LC tested on agar and it was healthy. They are chilling in a mini fridge at around 38-40 degrees F.

Was this a bad move? i was reading (before the shroomery) that this was a decent 'short term' (6 months - 1 year) way to store LC an keep cultures around.

My long term storage is the Castellani Method with agar wedges in distilled water at sub 40 degrees to put the culture into stasis.

Does this track or did i just do a whole lot of pointless work ?

I don't store LC so I don't know about that, but I keep my cryovials at room temperature, I don't want them going in stasis because of temperature, I want them to go in stasis because there's no resources. And as far as I have read room temperature is fine. I have revived 2 year old cryovials without any problem at all.

I don't have a dedicated fridge for mush cult so temperature fluctuations would be a factor. I did store plates in the fridge when I first started out and most of it just went bad because of condensation dripping down on the mycelium :crankey:

Do you notice a difference in colonization speed from agar that has higher nutes?

Like let's say I germinated on a .5% or 1% lme plate, and transfer to a 4%. Would wedges from the 4% colonize in any way faster? Or no?

Edit: Also, can someone explain to me why honey is out of fashion? Have people run side-by-side tests and found malt extract is so much faster?

Personally I don't like sugary media for liquids or agar for that matter. Honey tends to produce a lot of sediment, this makes it harder to identify turbidity.

I'm also not a huge fan of Malt extract either. It also can still produce sediment and I also find it quite sugary. It's better than honey but not much IMO.

My go to for both agar and LC was always potato, instant potatoes are easier but leave a lot of sediment. But they are hands down a far better nute source IMO. GW water is probably my second choice and brf would be the 3rd place option for me.

Are you talking agar to agar transfers? If that's the case then no, the opposite if any. With high nutrient plate I get denser growth and thinner growth with low nute plates that seems to move a little faster, but not much.

I re-run. WOrst case they're too sticky and you have to toss, and you were already ready to do that, so nothing to lose.

Two main ways to run dry by accident:
* Not being all the way in "sealing" mode on the flipper mode
* Jars slghtly too tall and pushing the lid up so it can't seal fully

I can run 3 quarts and a pint, or a tall 750ml mason jar in my 8qt. It's important to leave the shortest for the back where the vent is.

Hippie, take out your silicone sealing ring inside of it, wash it, then put a neutral oil like canola on a paper towel, and relubricate the seal, stretching it gently across its circumference.

I am cautious since I run grain jars for 3 hours after 15 minutes venting in the IP, so I put closer to 4 cups. Grains being under the water line doesn't really influence anything in the end does it?

For sure.  I fill the water halfway up my jar sometimes in my Presto and never had any issues..

Another question.

Last night I prepared four half pint with Millet and the appropriate water for no prep. left them on the counter while the insta was running its current batch.

This morning when I look at the jars a lot of the water noticeably got absorbed.  so you can no longer swirl the seeds around in the jar.

These things are still good to go in the insta as is right?  I mean the water didn't go anywhere. And since it's still there. I don't need to add more right? Seems like common sense. Just trying to make sure my logic is on point here.

Yep they're fine but with NSNS it's best to smack em around as soon as they get out of the pot to minimize clumps while they're still mobile like that. I don't add any more water when I re-run

I highly doubt it, the only time I've read or heard about mycelium jumping faster from agar to grains is if you use grain water plates from the same grains as in your jar, and even then I wouldn't imagine it would be more then a couple of days.

It does seem like 1% is faster.

Is there even any reason to use 4% agar? Unless you were growing exotics or culinary species. Or maybe trying to pin on agar.

4% agar? Like 4% nutes in agar? If so, no. In my opinion anything over 2% is a waste and only makes it harder to view what's going on on the plate.
