Gymnopilus luteofolius "Hornsby's"
https://mushroomobserver.org/obs/574089
Quite an aggressive strain, here I have plain grain that colonized CV and I cased with with a bit of vermiculite, cactus soil, and H202 soaked poppy seeds. I'm thinking of messing with the casing by grabbing each seedling and repotting them. Idk. I sort of want to get fruits first.

I let Hornsby's dry out a little bit, by cracking the lid of a modified monotub, and it stayed like that throughout the night and into the noontime. The poppy sproutlings were wilting, not sure if due to lack of real lights/strong LED's, if thats the result of it drying out, or the lack of moving air and the weakness of the cotyledobs. Most likely it was due to the dehydration of the casing. I had already assumed the casing was dry and if not the casing, the substrate itself. I think my strategy here is to slowly re-introduce & adhere to what I believe to be the proper fruting conditions.
Taking into account of what I already know about the species, I am intending on getting my surface conditions back, and if I can be so bold as to press the envelope; I have introduced a second bubbler. My intentions is for it to within a few hours quickly achieve proper fruiting conditions of the casing and/or substrate to sort of "make up" for letting conditions get too dry. I also wanted to be careful of Gymnopilus' sensitive surface mycelium, while still giving the project the overall water I think it needs. With any luck, I should be seeing the pins within a few days now, and I get the added benefit of letting the subtropicalis setup get the humidity and fae it night need, especially since I just re-cased it. Hopefully they like the same conditions and fruit wonderfully together.
I'm not sure if it'll be overall more wet than just one airstone in a pint jar by itself, because the two airstones in the tub are connected to a single 10-30 gal tetra airpump; and one jar seems to bubble leds intensely to my eyes.

Yeah, doesn't look like there is anything to save in your case. Maybe try and obtain packs of swabs and bring them with you when you go hunting. Your season is still in full swing, you could still start this project over and end up with some healthy streak plates.

Honestly I’d rather not fuck around with dirty prints again haha, have a few laying around already if I really wanted to punish myself. πŸ€£πŸ˜‚
I’ve got a few spots that I monitor so I’m just gonna wait until I see some young, fresh pinsets and clone from there. We’ve had a good soaking rain over the last few days so hopefully I can find something soon.

Might have been fine this last go but I was a bad cultivator and didn’t have any plates ready to go. Took waaay to long for me to get my shit together and prep some agar so yeah. I’ll try not to blow my load over anything white and fuzzy so fast 😬

I think I've got my first successful culture! It's been growing pretty  slow, but it's big enough to get some decent pictures now. This plate is from my G. Luteus spore print.

I guess I'm wondering two things. Is this healthy mycelium like I think it is? And when should I make some transfer plates if it is? I can't recognize anything else living in this plate which is sort of shocking to me. The cap I printed was pretty dirty. That and my inexperience have me doubting myself. But it's not like any of the mold and bacteria I've been growing. And it's got that powdered sugar look that I read about here, so I'm holding out hope for now.

Well, I'm gonna be the one to dash your hopes, but it builds tenacity. Look at you, just a week ago you had your hopes dashed and have had to bounce back and done and excellent job. Where do you get your plates? Hop in my DM's man. A slow growing mycelium like that is I'm sorry to say, not even what you thought it could be.

Hoping for redemption and finally getting a clean culture of subspectibilis. On my way home from work I noticed one of my spots was producing, super stoked I caught them young.
2 years ago in this spot I harvested 4 1/2 pounds in a single flush! And that was only half 🫠

Also came across an old bamboo agar plate from March that was a T1 after an attempt at cabin sequestering. No obvious contamination but definitely some kind of growth happening there. T2 that as well, guess we’ll see what happens.

That definitely looks like Gymnopilus mycelium.  Im glad you got a clean culture, both thru cabin sequestration and with a nice, clean, young flush of subspectabilis. I hope your clones turn out good. I have had a rough go at cloning, but it's nothing I can't handle, and if spores are unavailable I sometimes will have good luck simply dragging it along the surface of the agar. Someone also told me you can straight dip tissue in iso alcohol and then take your segments of flesh for cloning.
Once punctifolius fruits for me, Id like to have a nice culture of that from a clone if I can get one off it. It seems to be repeatedly colonizing the casing layer, dying off, and then colonizing the casing layer again. I think it has done it three times and on this last time, I switched up the fruiting conditions and took it out of the tub with an airstone and into a plastic bag with micropore tape over the top. It seems to be looking healthy now.

Anyone else skipping the casing on Gymns?
I had more success growing them in a bag of rehydrated hwfp (no pasturizing or sterilizing) and then stripping the bag off at colonization. When I find them in the wild they're growing straight from wood chips.

When I tried do them in a tray with casing they contaminated.

I don't have a lot of experience with these so I'm interested in some other opinions here.
