for those of you who propogate mycelium in a liquid culture and use quart jars... how do you retrieve the mycelium once it's ready to be taken into a syringe???

i shake it up and suck it up with a syringe.

if the myc won't break up, a sterilized marble or something around that size does the trick.

zero, do you use quart jars? i've only ever used pint jars for liquid myc. I see all these pictures of quar jars but im afraid that i wont be able to get my needle all the way into the jar without causing contamination. any ideas?

I did my liquid culture in a tupperware container, i realised when it came time to fill up the syringe i wouldnt be able to reach the liquid. So i took the container into a clean environment and opened the lid completely and sucked it up like that. I am very unsterile and never seem to have problems, i guess i have been lucky.

Gently swirl for a couple minutes everday to ensure that the tissue will be managable for aspiration!

I drill a hole close to the bottom of the quart jar and plug it with an injection plug. Works beautifully!

Here's a couple pics







I used a 20 dollar dremel drill from wall mart with some diamond bits.
Takes a while. Have to run it on 5,000-10,000 rpm(the only settings) to avoid overheating and breaking the glass. Holding the jar under a stream of room temp water while drilling is a good idea too.

Throw in a couple pieces of broken glass, marbles whatever before sterilization. :thumbup:

Any lid that will PC, fit a quart jar, couple pieces of rubber tubing, drill, bit, polyfil & rtv high temp gasket sealer.



Or same with metal lid

simply throw some glass sheds in there, rotate for a min or two and draw
up through self healing silicone injection port.

Tubing method is better. Why? You have tyvek under the lid, which you must poke needle hole in to suck out solution. Tyvek gets damp & will WICK in contams.

With tubing, you can clamp off & keep filter DRY.

Ok, I just have to ask the dumbest question...
You guys are getting all fancy with it which is way cool and all, but why not tip the damn jar enough to bring the liquid ( assuming you have reduced the contents to a homogeneous slurry ) close to the lid ( where the needle can reach ) and extract from there?

dint think of that, but actually i did couple of cultures with my method,
did it three times to be precise and let them go for 2-3 weeks,
no contams :smile:

I agree with not using the tyvek, it doesnt seem to have any use in lc's. As for lids i havent tried agars tek but this works good for me. I never get the polyfill wet because i use small mouth jars here as Mr stamets recomends and i only fill them half way. I get about 150ml of lc which is plenty. i used to use 1/2 gallon jars but i just never got around to using all of it.
