Sorry for the worlds shittiest picture but I got growth 48 hours later. The black spot is ofc a dark spore streak

Last attempt I had 3 jars have a couple white spots appear after 72 hours, although 12 hours later they turned green/grey.

This attempt is also with a different print from a grow i did about 2 years ago. Also using Malt/yeast instead of potato/karo

Anyways here's to hoping it stays white, so far this is the first visible growth out of the 12 jars I made. I anticipate there will be more colonies that pop up, but leaning towards going with the fastest

24 hours later, still not 100% sure if myc, kinda wish I didnt dye the agar now, hard to see anything through the jar. only 1 other jar has growth, about the size as the previous Pic, but 10 others do not have any visible growth

Almost out of petri dishes. Thinking about using the small cups instead. Does anyone have a link of where to get the cups?

Petris allow you to see what's going on much better than the cups.

I just acquired some PP5 cups thinking I would be able to reuse them and they are prepour. They had so much condensation! I put them back in the SAB and used a paper towel to remove all the condensate and now hoping they aren't contaminated. I will be purchasing more regular petri dishes so I can see better as well. Just my 2 cents.

Unless your paper towels were sterilized, you def just ruined those cups.

I assume you're stacking them during sterilization? Use the bottom ones that dont get condensation and loosen the lids on the others and let them dry.

:whathesaid:

Even with sterilized paper towel there is still a chance for contamination.

Condensation is from pouring too hot and happens to regular petris too

My agar plates are always lack luster. These are all T3s from spore, with the acception of a spore plate in pic I think 5. Is it just a matter of making more transfers until I get rhizo growth?

Are these all cubes? A lot of other species mycelium will not express rhizo at all.

As for cubes, it depends, they can switch expression on the plate, they might go rhizo if you transfer to a low nutrient plate or a higher nutrient plate or if you use a different nutrient source i.e. you transfer from MEA to PDA.

Rhizo does not really mean anything except that a culture expresses rhizo and it's easier to distinguish mushroom mycelium from mold mycelium but that is about it.

You can grow canopies of potent mushrooms from either tomentose or rhizomorphic, or cultures that express as both!

What's important is that the culture is clean

EPIC!

I did throw a pan, tamp, sub and semp in there.
So all cubes express rhizo, pans don't. Do other Ps species express rhizo?

All cubes can express rhizo, depending on the genetics some varieties are more likely to than others.

Pans, tamps (section mexicanae) and semps (section zapotecorum) will not but subs (section cyanescens) can.

Nothing in the Panaeolus genus will express rhizomorphic and as for the Psiocybe genus I think species in section aztecorum, caerulescens, cubensae and cyanescens all can but I am not sure if it's all species in those sections, and maybe I missed a section?

Epic. I feel like my mycelium is hella wispy and too transparent. Maybe I'm paranoid maybe I have contam. Either way I might have a protocol where I put all my cultures through a acbin sequestering procedure.

When looking at those photos, can you, as an experienced cultivator tell what areas I should be taking for T4s?

Thanks for your helps as usual I

Whatever the species I'm working with, I'm looking for fast organised growth with a clearly defined leading edge, assuming you want fruit and not sclerotia when working with species in section mexicanae, I will avoid the mycelium with a pigment and select for the white stuff.

It's a little difficult when I don't know exactly which species each plate is and from only a single photo without a backlight but I will do my best

Plate 1:

I am assuming this is a cube.

If those spots are sediment, you could probably let this culture to continue to grow out and send it to grain, assuming it continues to grow organised with a clearly defined leading edge.

If you want to transfer, everything from about 1 to 8 o'clock looks good.

Plate 2:

This looks like semps? Or is it the Pan you mentioned?

It's really hard to see what's going on but I think the bottom culture looks better, 11 to 4 o'clock, hold it up to a back light and check the growth is organised etc.

Plate 3:

Assuming semps again? Maybe this is a pan?

I don't like how it's growing, possibly has bacteria riding along, if you want to try save it, give cabin sequesting a try, growth at about 9 o'clock looks the best I think.

Plate 4:

Looks section zapotecorum. Semps

Looks like you have an orange dot close to the transfer, 7 to 11 o'clock looks like the nicest growth.

Plate 5:

This looks like your germination plate so I will assume I should ignore this one.

Plate 6:

Maybe this one is the sub?

This looks sketchy, you might want to give cabin sequestering a try, looks like some nice growth between 12 to 1 o'clock and about 8 to 9 o'clock.

Plate 7:

These next plates all look section mexicanae so I'll assume they are tampanensis since you mentioned tamps.

Top culture, I cant see what's under the glad wrap but I don't like much of what I can see, maybe around the 3 o'clock region.

Right-hand culture, looks like 4 to 5 o'clock has some nice growth.

Bottom culture, this is the best looking culture on the plate, 4 to 10 o'clock section looks good.

Left-hand culture, section between 8 to 9 o'clock.

Plate 8:

Looks like another possible bacteria riding along culture, again cabin sequesting if you want to try save it, about 10 o'clock I think.

Plate 9:

Again, it is hard to see what is going on where the glad wrap is covering the mycelium but these all look clean from here.

Maybe let them continue to grow out and if you feel the need to transfer, again hold it up to a back light, look for the fastest organised growth with a clearly defined leading edge and transfer from there.

I hope that helps! Good luck.

My bad :facepalm3:

I look at a lot of mycelium so I took it as an unlabelled plates challenge.
