Finally caved

Trying the vabiooth autoclaveable dishes..  figured I’d start small just in case I didn’t like it (I’m sure I will.. the possibilities are already running wild in my brain).  The first time was pretty uncomfortable tho but that might be because I poured the jars first and getting the lids and shit situated wasn’t fun but the actual plates went pretty smoothly but I completely didn’t even think about the surface being level.  When I started pouring and all the agar went to the top, I kinda freaked out   I’ll fix that tho

Big thanks to Milkboy and his thread and holding my hand a bit.. pretty much followed that to the T.  Stipe and his vids and psilicon and his Thread

Coolio. Does it look like a healthy plate? Not sure how tomentose these are meant to be

From what I can make out in the picture, it looks good. bisporus is very tomentose just like Pa. cyanescens. If you ever see a rhizo, it's not Panaeolus.

I actually did that once with some leftover agar in a tupperware haha. It got full of nasty shit, specially green mold which I think was trichoderma. That nasty mold is all around my place and usually infects my pf cakes before they can give a second flush.

That’s why the concept of enmeshed mold is so strange to me. The myc looks a lot like cube myc but “weird” and not like any other contam I’ve grown voluntarily or by accident

When trich sets in before a second flush it's usually because you I introduced it. When I run clean agar into pf cakes, I get multiple flushes. I make a habit of not wasting anything so I send my donor plates to brf cakes because of their tolerance to contams. I get 1 good flush by bottom watering after the dunk and roll.

Yeah I don't see anything bad but it looks more like original multispore streak plates rather than a transfer with a wedge.

This is a T4 from spore. I don't have a picture of my T3 right now, but here is the T2 of this culture line.

T3 looked better, slightly more rhizo and stronger, thicker threads. Just for comparison, this is a T3 from a different  culture line, but the T3 from which I transferred the blue plate in the previous post looked almost like this but a bit better.

I’m cleaning clones of my best pf cake flushes in brf pucks right now, I also have some cakes ready to harvest soon, would you recommend to just toss the line with the enmeshed mold and start again from those other options?

I've had recent transfers look worse going into T3 and T4.  I've read somewhere that Myc could grow bored with the medium and just start fizzling. I had some golden teachers that were thick ryzo according to my notes, but the T4 just looked tired and almost...whispy. MEA standard 2%

Put it to RGS and its reaching out, hoping the change in nutes will show a change.

You might be onto something. The problem could be my agar or my brf agar mix. This is myc from a plin I got from a clone of my original mss. It’s getting more and more rhizo as it grows through the puck while another plin from the same clone plate just looks wispy and sad in agar.

Faht getting into agar and using a FFU is actually fucking with my head. They are about to take agar work into another dimension. A dimension we cant even fathom and I am here for it

 thats too much pressure.  Not gonna try and reinvent the wheel here… I just wanna clean up my cultures and clone quickly

I’m not gonna dip my stem butt in peroxide before pulling it apart so I could clone in the flow without shit blowing all over my clean tissue and using a syringe that I bent the tiny needle tip in a little hook to scrape the smallest tissue piece off easily

Actually I did do those things lol. Idk how well it worked yet (except for the syringe scraper, that shit was fast af and it comes with a built-in water cooler)

I tried a scalpel at first but the tissue wouldn’t come off the blade easily and I had to wait for the damn thing to cool and I didn’t know if it was cool enough so it didn’t kill my tissue transfer.  Tissue was super easy to get off the needle… just twist it so the tip went in the agar but the tissue stayed

When I get a free month, I’ll read through this whole thread and see what tricks ppl are using

Did you bend space and time while you poured the plates?
Otherwise I'm disappointed.

I love the syringe scoop idea! Unless the scalpel is brand new (very sharp), it's hard to make clean cuts that want to come off of the mushroom and the scalpel. Did you use pliers to bend the needle top?

I'll bet it's pretty convenient for agar to agar transfers, too.

Another use of a syringe for cloning and cleanup transfers is the Josex poke, where you just poke into the inner mushroom tissue or agar plate and then squirt a few drops onto the new plate or BRF puck (pushing the plug of mycelium out of the needle).
