Re: AGAR ENVY! (Anything and All things agar!) [Re: Ben Phartin]

Hi guys can I get an expert’s opinion on this transfer? It’s a T4 from a GT MSS and it’s 6 days old.

It was taken from this plate which was the most “rhizo” growth I could get from that MSS.

I’m 99% sure it’s cube myc because I’ve used some plates as LI for pf cakes successfully. But after pouring some new agar plates with lower nutes and blue food coloring all my myc transfers started looking wispy again and a bit aerial. My first plates didn’t have the myc so “3D”, it grew stuck to the higher nute agar. I don’t know if the chinese plates I bought are bad, or if some bad sterile technique on my part introduced contams that are stressing the myc.

Anyone has any idea of why the myc could become wispy and aerial? I’m also getting some pp5 containers to transfer there and rule out contams from the plates.

Not an expert, but the mycelium on that transfer looks just fine to me. I can't tell from your picture if the nonuniformity elsewhere on the plate is bubbles, sediment, a non-solid background, or something else, but nothing about the look of the mycelium makes me think there's contamination.

Some cultures seem to struggle on certain media. Lots of people say it shouldn't matter, but I get much less wispy growth from my affected cultures with a bit of yeast (about 0.2%, nutritional or baker's, doesn't matter) in the agar mix. It will make sediment, which is possible to filter out or decant if you care (I don't).

Yeah not too bad at all I'm.gonna make a transfer or 4 n send the other plates of it and the rw , rw in my experience has always performed better on grain idk why bit it's always held true for me , bother time I transfer a Wedge to a plate and a jar at the same time the jars ready to shake days before my plates colinized

I have the B+ plates I showed a week ago for their two week checkup. They're still messy but I wanted the opinion of some experienced eyes. Learning goes faster that way.

I have 10 more plates of MEA available for transfers. The goal with this project is experience, and eventually getting a few clean plates for use. (And yes, primo plates have been added to the shopping list. Working with 30 plates to start has convinced me I want to continue and that I can budget in a bigger commitment.)

First, LC on MEA in HG. Pic is horrible. Focus is on the lid. I know. Just including this for completeness. They are as messy in person as they look in the picture. I'm thinking that from these two plates I'll take one transfer total. Incidentally these have started to eat the dye just a little.

Next, samples from a grain jar on the same plates. Left is messy like the above but looks much stronger. it got washed by a drop of condensation. Right colonized more from the center. Is that good rhizomorphic growth or more stressed? I'm thinking one sample from each of these. Left plate from some random edge. Right plate from the 9 or 10 o'clock position?

Last the water agar plates. I hope these are odd enough to be interesting. The dye really got eaten up on these. Guess that blue won't be useful. These all ended up similarly wispy. Again, one transfer from each of these as far from the start as it seems to have progressed? I'm wondering if they're all equally clean, equally dirty, or if there will be mixed results across the transfers.

That will leave me with 4 plates in reserve for further transfers but also collect a cross section of different sample conditions. I'd love to hear a sanity check on that plan or any insights on what growth is visible and I hope the water agar plates are novel enough to be interesting.

PS: The gourmet plates from the last post are boring enough that I'm confident in what they're doing and how useful they'll be. Going to dice up the best ones use a half plate each in some grain next week.

edit: 6 transfers done.

Ape plate! Still havent done a proper grow yet, my first shoebox just contammed. Think I should clone the big boy? It is already a clone culture, I think.

What would you do with this plate? A batch of master jars that each just have a plin?

How's this for Bisporus Washington?

I am new to this, and fortunately I know someone who has helped me have access to a flow hood and plates so I did 4 of them in a lab setting. 2 with a few drops of Albino Treasure Coast LC and 2 with Galactic Gorilla LC. The GG was so slow to take I thought it was a lost cause. One didn't make it, but I have 2 healthy ATC plates and one healthy GG plate to take from and propagate. Makes me happy to see them develop. Check em out.

Nice.

You know how they have those neighborhood "hacker spaces" where people can go work on projects, the place has tables, fast WiFi, coffee, a 3D printer, soldering irons, volt ohm meters, a drill set, etc?

Would be cool if one had a flow hood.

After a few days growth those little islands of growth should have expanded to be big enough where you may see sectoring occurring....

I think your use of "sectoring", if I'm understanding youm may be based on a misnomer that people used to say on here a 10 and 15 years ago (something from a Stamets book, haha), and people still claim on reddit a lot, but that it's not something TCs mention here now.

...the idea that you want to transfer repeatedly to get a "single isolated strain" or some such bullshit. ....which was disproven by the guy here who did 80 generations of transfers on PE and kept getting new sectors.

If you're not talking about that can you give more detail please? Thank you.

This first one looks good. I'd send it to grain. Someone else may say "one more transfer" but I'd send it.

The other ones I'd do another transfer but you're getting there.

Haha it's interesting you say 10 or 15 years because,  as you can notice from my post history,  that's precisely around the time I left for a long hiatus. And you're exactly right, the "sectoring" I referred to was meant to describe exactly what you mentioned. I haven't been around in forever and for sure am catching up on all kinds of updates and innovations that have transpired in the last decade or so. It's astonishing how much things evolve and how rapidly they change and progress. Pucks,  dyes, oats, shoeboxes... you name it. All the F1, T1, crossbred hybridized jargon is still foreign to me and I'm indeed struggling to keep up for now haha. But thanks for clearing that up for me. Good observation BTW. 👍

K sectoring is bullshit. Duly noted ✅️ 👌

How much did the transfer roll around? Those look like just spots where the transfer touched. Just a quick touch is enough to start spreading mycelium

Haha that'd be awesome! A little myco lab cafe. I love your set up there. I unfortunately don't the space nor the cake for a flow hood so I need a modestly sized SAB for home lab work.

Lol so wait,  after reading my use of the term, you're saying that is a correct understanding of the legitimate process known as "sectoring?" Just want to make sure I'm not confused still lol.
